Drp-1在糖尿病大鼠坐骨神经病变中的作用及白藜芦醇的干预影响

摘    要:探讨发动相关蛋白1(dynamin-related protein 1,Drp-1)在糖尿病大鼠坐骨神经中的作用及白藜芦醇干预的影响。采用链脲佐菌素(STZ)一次性腹腔注射诱导建立糖尿病大鼠模型;将制模成功的大鼠随机分为糖尿病对照组(DM组)、白藜芦醇低剂量治疗组(L治疗组)和白藜芦醇高剂量治疗组(H治疗组),另设正常组(N组)。应用Real-time PCR检测各组大鼠坐骨神经中Drp-1的表达水平,免疫组化检测各组Bax、Bcl-2、Caspase-3的蛋白表达,电镜下观察各组坐骨神经的超微结构。结果表明,超微结构显示DM组大鼠坐骨神经髓鞘疏松、板层排列紊乱、线粒体肿胀变性,出现溶解破碎;白藜芦醇治疗组上述病变程度较轻。DM组与N组相比,Drp-1、Bax、Caspase-3蛋白表达明显升高,Bcl-2蛋白表达明显下降;治疗组与DM组相比,Drp-1、Bax、Caspase-3蛋白表达明显下降,Bcl-2蛋白表达明显升高。Drp-1蛋白表达升高促进糖尿病大鼠细胞凋亡,坐骨神经变性;白藜芦醇能降低Drp-1表达,减少细胞凋亡,对糖尿病大鼠坐骨神经起一定的保护作用,且疗效跟剂量相关。
