穿心莲软胶囊与穿心莲片的药理作用及急性毒性实验研究

摘    要:【目的】观察穿心莲软胶囊的解热、抗炎作用及其急性毒性,并与传统剂型穿心莲片进行比较。【方法】采用细茵脂多糖(LPS)致家兔发热、啤酒酵母致大鼠发热动物模型,观察穿心莲软胶囊的解热作用;采用角又菜胶致大鼠足肿胀、二甲苯致小鼠耳肿胀和醋酸致小鼠腹腔毛细血管通透性增加的动物模型,观察穿心莲软胶囊的抗炎作用,并对小鼠进行该制剂的急性毒性试验。【结果】穿心莲软胶囊高、中剂量可抑制LPS引起的家兔发热(与模型对照组比较,P〈0.05或P〈0.01)。且与等剂量的穿心莲片比较具有显著性差异(P〈0.05);穿心莲软胶囊高、中剂量可降低啤酒酵母引起的大鼠直肠温度升高,降低角叉菜胶引起的大鼠足肿胀(与模型对照组比较,P〈0.05或P〈0.01),作用与穿心莲片相仿(P〉0.05),对二甲苯、醋酸介导的急性炎症渗出,穿心莲软胶囊高、中、低3个剂量均有抑制作用(与模型对照组比较,P〈0.01),其中中剂量组的抑制作用优于穿心莲片组(P〈0.05);急性毒性试验表明小鼠1日最大耐受量为110.25g/kg,相当于成人日用量的525倍。【结论】穿心莲软胶囊具有良好的解热、抗炎作用,部分药效指标优于传统剂型穿心莲片,且无明显毒副作用。

Abstract:Objective] To compare the antipyretic and anti-inflammatory actions and acute toxicity between Chuanxinlian Soft Capsules (CSC) and Chuanxinlian Tablets. Methods] Antipyretic action of CSC was observed on rabbit fever models induced by lipopolysaccharide (LPS) and rat fever models induced by baker's yeast. Rat models with pedal swelling induced by carrageenin, mouse models with auricle swelling induced by dimethylbenzene and mouse models with celiac capillary hyperpermeability induced by acetic acid were used to observe the anti-inflammatory action oi CSC. Its acute toxicity in mice was also evaluated. Results] High- and moderate-dose CSC reduced fever in rabbits induced by LPS (P < 0.05 or P < 0.01 compared with the model group) , which differed from that of the same dose of Chuanxinlian Tablets (P < 0.05) . High- and moderate-dose CSC reduced fever in rat fever models induced by baker's yeast and reduced the pedal swelling in rats induced by carrageenin (P < 0.05 or P < 0.01 compared with the model group), which was similar to that of Chuanxinlian Tablets (P > 0.05). High-, moderate- and low-dose CSC has an inhibitory effect on acute inflammatory exudation (P < 0.01 compared with the model group), and the effect of moderate-dose CSC was stronger than that of Chuanxinlian Tablets (P < 0.05). The results of acute toxicity test showed that the maximum tolerated dose to mice was 110.25g/kg, which was as much as 525 times of clinical dosage for each day. Conclusion] CSC has a remarkable antipyretic and anti-inflammatory actions and has no obviously toxic and side effects. Some of its pharmacodynamic parameters are superior to those of Chuanxinlian Tablets.
