双黄连治疗人巨细胞病毒小鼠肺炎的实验研究

摘    要:目的研究双黄连粉针剂对人巨细胞病毒(HCMV)感染的小鼠肺组织病理改变和病毒DNA载量的影响。方法取4周龄SPF级Balb/c小鼠60只随机分成6组,建立HCMV小鼠肺炎模型。然后分别进行双黄连及更昔洛韦腹腔注射治疗。用药10d后,处死小鼠,对小鼠肺组织进行电镜和光镜病理检查,同时用荧光定量PCR方法检测小鼠肺组织HCMV DNA载量。结果双黄连大、中剂量治疗组小鼠肺组织病理损害较感染模型组明显减轻,HCMV DNA载量分别为1628.80 copies/100mg和1869.37 copies/100mg,较感染模型组(468527.40 copies/100mg)有显著性降低(P<0.001),与更昔洛韦治疗组(1521.61 copies/100mg)相比差异无显著性(P>0.05),双黄连小剂量组小鼠肺组织病理损伤及DNA载量(462547.90 copies/100mg)与感染模型组相比差异无显著性(P>0.05);正常对照组肺脏未见异常改变。结论双黄连能明显减轻HCMV急性原发小鼠肺部炎症损害并降低HCMV感染小鼠肺组织中病毒DNA载量。

Abstract:Objective To investigate the effect of Shuanghuanglian(SHL) on the the lung of the mice infected by HCMV.Methods Sixty Balb/c mice aged 4 weeks were randomly divided into 6 groups.Murine model of HCMV pneumonia was established.The mice were treated with GCV or SHL i.p daily for 10 days and then sacrificed.The pathological change of lung tissue was examined by light and electron microscope.The load of lung HCMV DNA was detected by fluorescence quantitive PCR.Results The pathological change was less severe in high and middling dosage SHL group than HCMV model group.The load of lung HCMV DNA was significantly less in high and middling dosage SHL group(1628.80 copies/100 mg,1869.37 copies/100 mg respectively) than HCMV model group(468527.40 copies/100 mg) (P<0.01).No significant difference was discovered between GCV group(1521.61 copies/100 mg) and high,middling dosage SHL groups(P>0.05).The pathological change and the load of lung HCMV DNA in low dosage SHL group(462547.90 copies/100 mg) had no notable difference compared with HCMV model group.The lung tissue of control group had no abnormal change.Conclusion SHL can reduce the load of lung HCMV DNA and lighten lung histological pathology change in HCMV pneumonia model.
