宫颈鳞状细胞癌组织中Skp2与P27~(Kip1)的表达及其临床意义

摘    要:目的探讨细胞S相激酶相关蛋白(Skp2)和P27Kip1在宫颈鳞状细胞癌(SCC)发生过程中的表达及二者关系。方法用免疫组化方法检测10例正常宫颈组织、43例宫颈上皮内瘤变(CIN)和34例SCC组织中Skp2、P27Kip1蛋白的表达,用RT-PCR法对34例新鲜SCC组织进行Skp2 mRNA表达的检测。结果Skp2 mRNA在SCC呈过表达;Skp2蛋白表达在SCC发生过程中呈增高趋势;Skp2蛋白阳性率与淋巴结转移有关(P〈0.05);COX分析表明,Skp2是一个独立的预后参数。P27Kip1蛋白在正常宫颈中表达最高,随病变进展表达呈下降趋势;P27Kip1蛋白阳性率与淋巴结转移相关(P〈0.05);Skp2与P27Kip1表达呈显著负相关(P〈0.01)。结论Skp2高表达和P27Kip1失活致细胞增殖周期失调在SCC发生过程中起重要作用,可作为SCC早期诊断、临床生物学行为和预后评价的新的参考指标。

Expressions of Skp2 and P27^Kip1 in Squamous Cell Carcinoma of Cervix and Their Signifance

Abstract:Objective To investigate the expressions of Skp2 and P27^Kip1 in cervical carcinogenesis,and to discuss the relationship between the expression of Skp2 and P27^Kip1. Methods Immunohisto- chemistry was performed in the 10 normal cervical tissue and 43 CIN and 34 cervical carcinoma to evaluate the expression alteration of the Skp2,P27^Kip1 proteins.RT-PCR was performed in fresh normal cervical tissue and cervical carcinoma to detect the expression of Skp2 mRNA. Result RT-PCR analysis showed that Skp2 mRNA was overexpressed in carcinoma tissue. The positive rate of Skp2 showed an increasing tendency in cervical carcinogenesis. The expression of P27^Kip1 was the highest in normal cervical epithelium and tend to decrease with cervical diseases progression. Correlation existed between the expression of Skp2,P27^Kip1 and lymph node metastasis (P〈0.05). No significant relationships were found between the expression of Skp2,P27^Kip1 and other clinicopathological features,such as age,clinic grade,clinic stage,etc. The expression of P27^Kip1 correlated with Skp2 expression and it was negative correlation(P〈0.05). Conclusions Overexpression of Skp2 and inactivation of P27^Kip1 lead to disproportion of cell proliferation cycle,which play an important role in cervical carcinogenesis, and they might be considered as new prediction of early diagnosis, clinical biological behavior and prognosis of cervical carcinoma.
