布拉氏酵母菌对大鼠肠道菌群及血清炎性因子的干预研究

摘    要:目的 观察高脂饮食对大鼠肠道菌群及血清炎性因子的影响,探讨布拉氏酵母菌散剂在调节肠道菌群中的作用.方法 将36只大鼠随机分为A、B、C组,每组12只.A组喂养普通饲料,B组喂养高脂饲料,C组喂养高脂饲料,并予布拉氏酵母菌干预.实验初始、第10周末检测血清脂多糖(LPS)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及粪便中双歧杆菌、乳酸杆菌、拟杆菌、大肠杆菌、肠球菌数量.结果 3组初始时LPS、TNF-α、IL-6差异无统计学意义(P>0.05);10周末时B、C组LPS、TNF-α较初始时明显升高(P<0.05、0.01),A组无改变(P>0.05);B组IL-6较初始时升高(P<0.01),A、C组无改变(P>0.05);10周末时B组LPS、TNF-α、IL-6均高于A、C组(P<0.01),C组TNF-α高于A组(P<0.01).3组初始乳酸杆菌、双歧杆菌、肠球菌、拟杆菌、大肠杆菌比较差异无统计学意义(P>0.05);10周末时B、C组乳酸杆菌、双歧杆菌、肠球菌较初始时升高(P<0.01),A组无改变(P>0.05),B组低于A组及C组(P<0.01),C组低于A组(P<0.05);10周末时B组拟杆菌较初始时降低(P<0.01),A、C组无改变(P>0.05),B组低于A、C组(P<0.01);10周末时B组大肠杆菌较初始时升高(P<0.01),A组无改变(P>0.05),C组降低(P<0.05),B组高于A、C组(P<0.01),A组高于C组(P<0.05).结论 高脂饮食喂养会增加大鼠血清炎性因子释放,改变肠道菌群结构;布拉氏酵母菌可稳定肠道菌群结构,减少炎性因子释放.

A Study on Saccharomyces Boulardii in Intervention of Intestinal Flora and Inflammatory Factors of Rats
Abstract:Objective To observe influence of high-fat diet on intestinal flora and inflammatory factors of rats, and to investigate effect of Saccharomyces Boulardii powders in regulation of intestinal flora. Methods A total of 36 rats were randomly divided into group A, B and C ( n=12 for each group) . Group A was given normal diet. Group B was given high-fat diet. Group C was given high-fat diet and Saccharomyces boulardii powder. Lipopolysaccharide (LPS), interleukin-6 (IL-6) and tumor necrosis factor-α(TNF-α) in serum and numbers of bacillus bifidus, lactobacilli, bacte-roides, escherichia coli and enterococcus in feces at the beginning and at the end of 10 th weeks of experiment were detec-ted. Results There were no significant differences in LPS, TNF-αand IL-6 levels at the beginning experiment among 3 groups (P>0. 05). At the end of 10th weeks of experiment, LPS and TNF-αlevels in group B and C were significantly increased (P<0. 05, P<0. 01), while the levels showed no changes in group A (P>0. 05);IL-6 level in group B was significantly increased (P<0. 01), while the levels showed no changes in group A and C compared with those at the be-ginning experiment (P>0. 05). Levels of LPS, TNF-αand IL-6 in group B were significantly higher than those in group A and C (P<0. 01), TNF-αlevel in group C was significantly higher than that in group A at the end of 10th weeks of ex-periment ( P<0. 01 ) . There were no significant differences in numbers of fecal bacillus bifidus, lactobacilli, bacte-roides, escherichia coli and enterococcus at the beginning experiment among 3 groups ( P>0. 05 ) . At the end of 10 th weeks of experiment, numbers of fecal lactobacilli, bacillus bifidus and escherichia coli in group B and C were signifi-cantly higher (P<0. 01), while the numbers showed no changes in group A compared with those at the beginning experi-ment (P>0. 05);the numbers in group B were significantly lower than those in group A and C (P<0. 01), and the numbers in group C were lower than those in group A (P<0. 05). At the end of 10th weeks of experiment, number of fe-cal bacteroides in group B was lower (P<0. 01), while the numbers showed no changes in group A and C compared with those at the beginning experiment (P>0. 05), and the number in group B was significantly lower than those in group A and C (P<0. 01);the number of fecal escherichia coli in group B was significantly higher (P<0. 01), while the num-ber in group A showed no change (P>0. 05), but the number in group C was significantly lower compared with those at the beginning experiment (P<0. 05);the number of fecal escherichia coli in group B was higher than those in group A and C (P<0. 01), and the number in group A was higher than that in group C (P<0. 05). Conclusion High-fat diet may increase release of inflammatory factors and change intestinal flora of rats. Saccharomyces boulardii can stabilize in-testinal flora and reduce release of inflammatory factors.
