复方竹叶石膏颗粒对家兔急性放射性食管炎COX-2与MMP-2mRNA及蛋白表达的影响

摘    要:目的 探讨复方竹叶石膏颗粒对家兔急性放射性食管炎环氧化酶-2(COX-2)与基质金属蛋白酶-2(MMP-2)mRNA及蛋白表达的影响.方法 将40只雄性家兔随机分为空白对照组、单纯照射组、复方竹叶石膏颗粒组、康复新液组,每组10只.空白对照组不做任何处理,其余3组均给予6 mV-X射线对家兔食管单次照射32 Gy.给药组于照射前1周给药至照射结束后7d,4组均于照射后第7天留取家兔全长食管,检测COX-2与MMP-2 mRNA及蛋白表达情况.结果 单纯照射组中COX-2与MMP-2 mRNA与蛋白水平高于空白对照组,复方竹叶石膏颗粒组和康复新液组均低于单纯照射组,且复方竹叶石膏颗粒组低于康复新液组(P<0.05).结论 复方竹叶石膏颗粒能降低COX-2与MMP-2 mRNA与蛋白表达水平,从而发挥对急性放射性食管炎的防治作用.

Abstract:Objective To investigate effects of Compound Zhuye Shigao Granule (CZYG) on mRNA and protein expressions of cycloxygenase-2 (COX-2) and matrix metalloproteinases-2 (MMP-2) in treatment of rabbits with radiation esophagitis.Methods A total of 40 male rabbits were randomly divided into control group (n =10,group A),simple radiation group (n =10,group B),CZYG group (n =10,group C) and Kangfuxinye group (n =10,group D).All rabbits except those in control group were irradiated by 6MV X-ray at a dose of 32Gy for one time.The drugs were given starting from 1 week before the irradiation to at 7thd after irradiation in group C and D.Samples of the total length of rabbits'esophagus were collected at 7thd after irradiation in all groups to detect levels of mRNA and protein expressions of COX-2 and MMP-2.Results In group B,mRNA and protein expressions of COX-2 and MMP-2 were significantly increased compared with those in group A (P < 0.05),While the expressions in group C and D were significantly lower than those in group B,and the expressions in group C were significantly lower than those in group D (P < 0.05).Conclusion Compound Zhuye Shigao Granule can prevent radiation esophagitis by decreasing mRNA and protein expressions of COX-2 and MMP-2.
