摘    要:目的探索一种适合临床使用检测肺炎支原体(MP)的测定方法。方法采用实时荧光定量聚合酶链反应(PCR)技术分别测定漱口液、咽拭子的MP。结果在28例培养或补体结合试验(CFT)阳性的标本中漱口液阳性率为89.3%(25/28),咽拭子阳性率为71.4%(20/28),二者比较差异有统计学意义(P〈0.05);培养或CFT阴性的34例咳嗽、发热患儿的漱口液和咽拭子的MP均为阴性。结论采用实时荧光定量PCR技术测定漱口液中MP可以代替培养法,可作为诊断MP感染的依据。

Abstract:Objective To develop a rapid method suitable for quantitatively measuring mycoplasma pneumoniae (MP) in clinic. Methods Real time fluorescent quantitation polymerase chain reaction (FQ-PCR) was applied to measuring MP in gargle and pharynx swab specimens. Results Twenty-seven specimens were culture or complement fixation test (CFT) positive,in which the MP positive rate was 89.3% (25/28)for gargle and 71.4% (20/28) for pharynx swab. The difference of MP positive rate was statistically significant between the two kinds of specimen (P〈0.05). In 34 patients with culture or CFT negative,DNA of MP in both liquid of gargle and pharynx swab was negative, Conclusion FQ-PCR assay can substitute cultivation for detecting MP in liquid of gargle,which is used as a technique for diagnosis of MP infection.
