摘    要:对75株酵母菌经初筛、复筛和单倍体分离,得到胞内还原型谷胱甘肽(GSH)含量较高的单倍体酵母菌株S1(a),并进行遗传标记.经硫酸二乙酯诱变、紫外诱变及两者的复合诱变,ZnCl2、乙硫氨酸抗性交替筛选得GSH含量较高的单倍体F4(a,Arg-,ZnCl2r,Ethr),GSH含量与产量分别比出发单倍体提高了73.5%和118.1%.

Abstract:A yeast haploid S1(a) with high intracellular content of reduced glutathione (GSH) was selected from 75 yeast strains by primary screening and isolation of haploid, and then marked with auxotroph. The high GSH producing strain F4 (a, Arg-, ZnCl2 r, Ethr) was constructed by mutagenizing S1(a) with diethyl sulphate(DES), UV and UV-DES, followed by resistance selection with ZnCl2 and L-ethionine. The intracellular content and the yield of GSH by haploid F4 were increased by 73.5% and 118.1% over the original strain, respectively.
