4种肝脏吸虫蛋白质圆盘电泳和等电聚焦的比较分析

摘    要:应用聚丙烯酰胺凝胶圆盘电泳和薄层等电聚焦分析比较4种肝脏吸虫成虫可溶性蛋白质,结果显示肝片形吸虫、华枝睾吸虫、东方次睾吸虫和台湾次睾吸虫圆盘电泳的区带数分别为20、18、18和15条,主带数分别为5.4、8和5条;等电聚焦的区带数分别为28、22、24和24条,主带数分别为10、3、7和7条。蛋白电泳结果表明东方次睾吸虫和台湾次睾吸虫为两个独立虫种。用数量分类学的相似率表示4种吸虫蛋白电泳谱的相似程度,结果显示东方次睾吸虫和台湾次睾吸虫最为相似,亲缘关系最近。

Abstract:soluble proteins from the liver flukes of four species of adult worm were analyzed using disc elec-trophoresis (PAGE) and isoelectric focusing (IEF). The numbers of bands distinguished by PAGE for Fasciola hepatica,Cloworchis sinensis,Metorchis orientalis and M. taiwanensis were 20,18,18 and 15 respectively and the numbers of prominent bands were 5,4,8 and 5 respectively;the numbers of bands distinguished by IEF were 28,22,24 and 24 respectively and the numbers of prominent bands were 10,3, 7 and 7 respectively. It showed that Mo and Mt are two valid species in electrophorepic patterns A method of numerical taxonomy, percentage of similarity, was used to express the degree of similarity in protein paterns of four different species. It also showed that Mo and Mt were the most similar and their relationship was nearest.
