激光间质内热疗联合顺铂治疗大鼠脑胶质瘤的实验研究

摘    要:目的 评价激光间质内热疗(ILTT)联合顺铂治疗大鼠脑恶性胶质瘤的效果。方法 SD大鼠64只,尾状核接种C6鼠胶质瘤细胞14天后,随机分为对照组(不接受治疗)、顺铂治疗组、ILTT组、ILTT联合顺铂治疗组,每组16只。每组中10只留作观察生存时间(观察期40天),6只于治疗后第7天测量肿瘤最大横径。顺铂治疗时大鼠尾静脉注射顺铂1μg/g,2天1次,共计3次。ILTT治疗时将Nd∶YAG激光经光导纤维导入肿瘤区,激光波长1.06μm,功率1.5W,时间120s,间隔120s,共计2次。结果 (1)大鼠平均生存时间对照组26.1d±3.6d,顺铂组27.3d±3.9d,ILTT组30.4d±5.3d,联合治疗组34.4d±4.0d。联合治疗组与对照组及顺铂组比较,差异有显著意义(P<0.05)。(2)肿瘤最大横径对照组6.2mm±0.2mm,顺铂组6.2mm±0.1mm,ILTT组4.8mm±0.2mm,联合治疗组4.9mm±0.1mm。结论 激光间质内热疗联合顺铂治疗对大鼠脑胶质瘤有较强的抗肿瘤效果。

Abstract:Objective To evaluate the therapeutic effects of interstitial laser thermotherapy (ILTT) combined with cisplatin chemotherapy on rat glioma model with deeply sited tumors.Methods C6 glioma cells were injected into the nucleus caudatus by stereotactic technique to induce transplanted gliomata in the rat brains. The rats bearing gliomata were treated with ILTT in combination with cisplatin. The survival time of rats was observed up to 40 days after ILTT, and the tumor size of rats at 7th day after ILTT was measured. The data in different groups were statistically analysed.Results The mean survival period of rats was 26.1 d±3.6 d in the control group, 27.3 d±3.9 d in the cisplatin treated alone group, 30.4 d±5.3 d in the ILTT alone group and 34.4 d±4.0 d in the ILTT combined with cisplatin chemotherapy group. At the 7th postoperative day, the maximum diameter of tumors measured in coronal sections of the brains were different between control group and experimental groups: 6.2 mm±0.2 mm of the control group , 6.2 mm±0.1 mm of the cisplatin treated alone groups, 4.8 mm±0.2 mm of ILTT group and 4.9 mm±0.1 mm of the ILTT combined with cisplatin chemotherapy group.Conclusions Based on the above described results, we would conclude that the combination of ILTT and cisplatin chemotherapy might provide a significantly greater antitumor effect in the treatment of glioma.
