血源性脑及脑膜转移瘤中VEGF表达及磁共振检测

摘    要:目的:探讨血源性脑及脑膜转移瘤中VEGF的表达水平及MRI检测意义。材料和方法:38只新西兰大白兔随机分为三组,分别从颈总动脉行血脑屏障开放接种VX2癌细胞,单纯接种VX2癌细胞及单纯注射生理盐水。接种后1周开始在不同时间行MRI检查。病理取材HE染色光镜下观察,免疫组化方法检测脑及脑膜转移瘤中的VEGF表达水平。结果:实验组脑膜成瘤率72%,脑内成瘤率62%。MRI表现为T1WI等信号,T2WI低或高信号。注射Gd-TDPA后T1WI有明显的增强效应。VEGF检测证实血源性脑及脑膜转移瘤明显高于正常组。结论:VEGF对脑及脑膜转移瘤的形成可能起重要作用,进一步研究VEGF抗体及其拮抗剂阻止或抑制脑及脑膜转移瘤是肿瘤治疗的发展方向。

Abstract:Purpose:To detect VEGF expression and MRI significance of hematogenous cerebral and meningeal metastasis.Materials and methods:38 New Zealand rabbits were divided into 3 groups.The first two groups inoculated with VX2 carcinoma from common carotid arterial with or without opening up blood-brain barrier(BBB) respectively.The last group was inoculated with physiological saline only.MRI including plan scan and enhanced scan was done at different times 1 week after inoculation.Pathological study were done under microscope by HE,VEGF expression was detected by immunohistochemical stains.Results:The rate of growth was 72%for meningeal metastasis,and 62% for cerebral metastasis.MRI showed the tumor appeared iso-intense on T1WI,hypo-or hyper-intense on T2WI with marked enhancement after intravenous injection Gd-DTPA.VEGF expression of cerebral and meningeal metastasis was higher than that of normal.Conclusion:VEGF may play an important role in cerebral and meningeal metastasis growth.It is a further direction to study VEGF antibody and antagonist in blocking or inhabiting cerebral and meningeal metastasis.
