胆囊收缩素对胆管癌细胞凋亡的调节作用及机制

摘    要:目的 探讨胆囊收缩素(CCK)对胆管癌细胞凋亡的调节作用及机制。方法 采用TUNEL染色、定量RT-PCR和反义寡核苷酸技术分别研究CCK-8S对QBC939人胆管癌细胞系诱发性凋亡指数(AI)和bcl-2/bax mRNA表达的影响、以及bcl-2在CCK抑制凋亡中的作用。结果CCK-8S组 AI显著降低、bcl-2mRNA表达显著增强,同时加入CCK受体拮抗剂L364.718或L365.206可拮抗;反义bcl-2寡核苷酸转染能逆转CCK-8S对凋亡的抑制作用。结论CCK能提高凋亡阈值、抑制胆管癌细胞凋亡,其机制与上调bcl-2基因表达有关。

Abstract:Objective To explore the effects of CCK on bcl - 2 - associated apoptotic pathways in cholangiocar-cinoma. Methods TUNEL technique and quantitative RT- PCR were used to examine the effects of CCK- 8S on the beauvericin - induced apoptotic index (AI) and expressions of bcl - 2 and bax rnRNAs, and antisense oligodeoxynu-cleotide (ODN) technology was applied to investigate the role of bcl - 2 in the CCK - mediated inhibition of apoptosis in human cholangiocarcinoma cell line QBC939. Results The AI and bcl - 2 mRNA expression significantly decreased and increased respectively in the cells exposed to CCK - 8S compared with those of control cells, and these two actions of CCK could be blocked by CCK receptor antagonist 1364, 718 or L365, 260. Furthermore, the inhibitory effect of CCK on apoptosis could also be abolished by bcl - 2 antisense ODN. Conclusions CCK is able to increase the threshold for apoptosis by upregulating bcl - 2 gene expression in human cholangiocarcinoma cells.
