| 摘 要： | 目的:探讨长链非编码RNA Gas5(lncRNA Gas5)在原发性肝癌组织中的表达及其对Hep3B细胞的侵袭作用。方法:采用RT-PCR检测50例肝癌患者肿瘤和癌旁组织中lncRNA Gas5的表达水平,分析其与临床病理特征的相关性;通过在Hep3B细胞中过表达lncRNA Gas5,利用Transwell小室检测过表达lncRNA Gas5后肿瘤细胞侵袭变化,RT-PCR和Western blot检测PTEN蛋白表达变化。结果:肝癌组织中的lncRNA Gas5表达水平较癌旁组织显著降低(t=9.759,P<0.01),并与肝门淋巴结转移(t=7.748,P<0.001)和肿瘤TNM分期(t=2.489,P=0.016)相关;过表达lncRNA Gas5后的Hep3B细胞侵袭能力减弱(P<0.01),PTEN基因和蛋白表达水平升高(P<0.05)。结论:lncRNA Gas5在肝癌组织中的表达下调增强肝癌细胞的侵袭能力,可能成为原发性肝癌术后判断复发的分子标志物。
