聚合酶链式反应诊断2型猪链球菌

摘    要:目的从分子水平鉴定四川省2005年不明原因疾病病原菌的属、种、型及毒力基因,为诊断和治疗提供科学依据。方法聚合酶链式反应(PCR)检测链球菌属特异性基因(TUF)、猪链球菌种特异性基因(Species)、2型和毒力特异性基因(CPS2J)以及毒力基因溶菌酶释放相关蛋白基因(MRP)和溶血素基因(SLY);同时与VITEK2等全自动生化鉴定仪鉴定结果进行比对。结果所试103份标本中,98份均具有所检测的5个基因,确定为猪链球菌2型;另5份标本排除猪链球菌可能。73株纯培养细菌用VITEK2全自动生化鉴定仪鉴定有68株菌被鉴定为猪链球菌2型,3株菌被鉴定为猪链球菌1型,2株菌不能鉴定。结论2005年四川省不明原因疾病疫情为猪链球菌2型感染人造成。PCR检测5个特异基因鉴定猪链球菌2型,其结果优于VITEK2全自动生化鉴定仪。

Abstract:Objective To identify the genus, species, type and toxic gene of pathogenic bacteria causing disease outbreak in Sichuan in 2005,in order to provide scientific basis for diagnosis and treatment. Methods Streptococcus genus specific gene (TUF), Streptococcus suis species specific gene (Species), S. suis serotype 2 type specific and pathogenicity specific gene (CPS 2J), and two toxic genes: Muramidase-released protein gene (MRP) and hemolysin gene (SLY)were detected by PCR,and the results were compared with the results of VITEK2 auto biochemistry identifier. Results Totally 98 of 103 strains tested were identified as S. suis serotype 2 by PCR because 5 genes of them were positive. The other five were not S. suis serotype 2.VITEK2 auto biochemistry identifier were used to analyze 73 strains,and 68 strains were identified as S. suis serotype 2, 3 strains identified as S. suis serotype 1 and the other two ones couldn't be identified. Conclusions The disease outbroke in Sichuan in 2005 was caused by Streptococcus suis serotype 2 which infected human beings. PCR based on testing five specific genes was more specific and sensitive than VITEK2 in S. suis serotype 2 identification.
