蒲黄、丹参对纤维蛋白损伤内皮细胞的保护作用

摘    要:应用传代培养的牛和人主动脉内皮细胞及原代培养的人脐静脉内皮细胞为实验模型,用1%纤维蛋白的琼脂薄膜复盖于融合成单层的细胞上,观察比较蒲黄、丹参对纤维蛋白损伤牛和人血管内皮细胞的影响。发现牛和人主动脉或脐静脉内皮细胞接触含纤维蛋白的琼脂薄膜后,部分细胞出现收缩移动和细胞间隙增宽,随后大多细胞脱落,少数聚集成团;而加入蒲黄血清或丹参针液的防治组,大部分细胞形态基本正常。加入蒲黄、丹参防治组的培养液中,LDH及ACP含量均明显低于实验对照组。提示蒲黄、丹参均能减轻纤维蛋白凝块对血管内皮的损伤作用。

Protective Effects of Pollen Typhae and Radix Salviae Miltiorrhizae against the Injurious Action of Fibrin on Endothelial Cells

Abstract:The protective effects of Pollen Typhae and Radix Salviae Miltiorrhizae against the injurious aetion of fibrin were studied in cultured endothelial cells including bovine and human aortic endothelial cells (BAEC 5-6th passage, HAEC 9th passage) and human umbilical endothelial cells (primary culture). They were divided into three groups. All of them were contacted with 1% fibrin in agar but two groups were treated with Pollen Typae and Radix Salviae Mlitiorrhizae respectively, the other served as the control group. In the control group contacting with fibrin 6 hours later the endothelial cells showed partial contraction, migration and the gaps between these cells being widened. After 24 hours a large number of cells were denuded and some aggregated in clumps. Nevertheless, in the test groups the cells were almost normal in shape, only a very few cells detached. The contents of lactic acid dehydrogenase(LDH)and acid phosphatase (ACP) in the medium of control group were significantly higer than in the test groups and the cell protein in controlgroup is much less than that in groups treated with Chinese herbal medicine. If the cultured cells were injured more seriously, the enzyme such as LDH and ACP releasing from these ceils showed significantly increased but the cultured cell protein decreased due to a large number of cells detached.These results also showed the morphological and metabolic changes in tbe injured cells were correlated.
