珍珠梅提取物对大鼠慢性肝损伤的保护作用

摘    要:目的:观察珍珠梅提取物对大鼠慢性肝损伤的保护作用.方法:72只大鼠随机分为正常组、模型组、秋水仙碱组和珍珠梅给药组,秋水仙碱和珍珠梅以灌胃的方式给药.对比观察秋水仙碱和珍珠梅对二甲基亚硝胺(DMN)所致慢性肝损伤大鼠的肝功能、肝纤维化、肝病理学及肝组织氧化指标的影响.结果:珍珠梅给药组大鼠肝功能指标丙氨酸氨基转移酶(ALT)、天门冬氨酸转移酶(AST)、白蛋白(ALb)、碱性磷酸酶(AKP)较模型组明显改善(P<0.05或P<0.01);肝纤维化血清学指标透明质酸(HA)、Ⅲ型前胶原(PCⅢ)和肝组织羟脯氨酸(Hyp)含量明显低于模型组(P<0.05或P<0.01);肝组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性明显高于模型组,丙二醛(MDA)含量较模型组明显降低(P<0.05或P<0.01);HE染色及α-鼠抗平滑肌肌动蛋白(α-SMA)的表达示肝组织纤维化程度较模型组明显减轻(P<0.05);珍珠梅提取物抗肝纤维化综合效果优于秋水仙碱.结论:珍珠梅提取物对慢性肝损伤有一定的保护作用,抗氧化作用可能是其主要作用机制之一.

Abstract:OBJECTIVE: To study the effects of Sorbaria sorbifolia extract (SSE) on DMN-induced chronic liver damage in rats. METHODS: Seventy-two rats were divided into four groups, normal, model, SSE and colchicin group. Serum hyaluronic (HA) and type precollagen (PCIII) were detected by radioimmunoassay. Degrees of fibrosis and alpha-smooth muscle actin (alpha-SMA) were determined by histopathology under microscope. Hydroxyproline (Hyp) and the markers of liver function were examined also. RESULTS: The levels of serum ALT, AST, ALb, AKP in SSE groups were notably improved as compared with the model group. Serum HA, PCIII and Hyp of hepatic tissue in SSE groups were obviously lower than those of model group. The markers of oxidative stress( SOD, MDA, GSH-Px) were strikingly improved also in SSE groups. HE stains and alpha-SMA expression showed that the degrees of liver fibrosis in SSE groups was slighter than that of model group. There was significant difference between the model group and SSE groups (P < 0.05 or P < 0.01). The rats in SSE groups were better than those of colchinicines group. CONCLUSION: SSE is able to protect against DMN-induced chronic liver damage, which may act through oxidative stress.
