基于UPLC-ESI-Q-TOF-MS的红禾麻提取物化学成分分析

摘    要:目的:建立超高效液相色谱-电喷雾-四级杆-飞行时间串联质谱联用技术(UPLC-ESI-Q-TOF-MS)研究红禾麻提取物中的主要化学成分。方法:采用Agilent Eclipse Plus C_(18)RRHD色谱柱(2.1 mm×100 mm,1.8μm);以0.1%甲酸水溶液-0.1%甲酸乙腈溶液为流动相进行梯度洗脱;流速0.25 m L·min~(-1),柱温40℃,进样量1μL。采用电喷雾离子源(ESI),负离子模式进行检测,运用Data Analysis软件,并结合保留时间、精确相对分子质量和相关文献进行数据分析。结果:通过结合对照品及文献报道的质谱数据对红禾麻提取物中化学成分进行分析,共分析鉴定13个化学成分,包括4个儿茶素类,3个苯丙素类和6个黄酮类,其中新绿原酸、隐绿原酸、绿原酸、芦丁、异槲皮苷、木犀草苷、山柰酚-3-O-芸香糖苷、紫云英苷和槲皮苷均首次在该植物中发现。结论:研究中建立的检测方法简便、准确,为快速鉴定分析化学成分提供一种有效的分析方法,并比较全面地阐明了红禾麻提取物中的化学成分,为其质量控制和药效物质基础研究奠定一定的基础。
