正常大鼠吸入一氧化氮肾脏抗氧化能力的变化

摘    要:目的:观察吸入外源性一氧化氮(NO)对正常大鼠肾脏抗氧化能力的影响,旨在评价其对肾脏的毒副作用。方法:72只大鼠随机分为3组,均行气管切开及呼吸机辅助呼吸,吸氧组(G_1)单纯吸氧,低浓度NO吸入组(G_2)和高浓度NO吸入组(G_3),除吸氧外,分别吸入40ppm和80ppm的NO。分别取6只大鼠建立正常值。数据行多个样本均数间方差分析。结果:血清中TAOC G_2与G_3组均高于G_1(P<0.01);G_2组MDA/SOD低于G_1和G_3(P<0.01)。肾组织中SOD三组变化不显著,G_3组MDA明显高于G_1和G_2组(P<0.01),G_2组低于G_1(P<0.05);三组ATP均低于正常值(P<0.01),但G_2明显高于G_1和G_3组。结论:吸入低浓度NO能不同程度的提高肾组织能量水平和抗氧化能力,而高浓度的NO则会有一定损伤作用。

Abstract:Objective:To observe the changes of anti-oxidation after inhaling nitric oxide to the normal rat' s kidney tissue. Methods: 72 rats were divided into three groups, All of the three groups tracheotomy were performed and intubation and mechanical ventilation were performed. The control group (G1)breathed O2(FiO2 = 0.40); Group of inhaling lower dense NO performed (G2) and group of inhaling high dense NO(G3)breathed 40ppm and 80ppm NO respectively besides greathed O2(FiO2 = 0.40) .In addition,6 animals served as normal controls for histological comparison and to set up the normal value ( n = 6). The data was analyzed in use of ANOVA for multiple mean comparison. Results: The level of TAOC in serum of both G2 and G3 were higher than G1 ( P < 0.01); The level of MDA/SOD in serum of G2 were lower than G1 and G3(P<0.01). There were no significant difference of the content of SOD in kidney tissue among three groups. The content of MDA of G3 in kidney tissue was higher than that of G1 and G2 was also lower than normal value ( P < 0.05); The level of ATP in tissue of three groups is lower than G] simultaneously ( P < 0.05); The level of ATP in tissue of three groups lower than normal value ( P < 0.01), but G2 was higher than both G3 and G1 ( P < 0.05). Conclusion: Inhaling lower dense NO can increase the energy level and antioxidation ability of rat' s kidney tissue in some degree, but high dense NO may increase the level of lipid peroxidation in rat's kidney in some degree. It suggest that inhaling lower dense NO is not harmful on rat' s kidney,but high dense NO was harmful on rat s kidney.
