熊胆冻干粉针剂对缺氧-再复氧损伤血管内皮细胞的保护作用

摘    要:目的探讨熊胆冻干粉针剂对缺氧-再复氧损伤的血管内皮细胞的保护作用及抗动脉粥样硬化机制。方法采用脐静脉血管内皮细胞(hum an umb ilical ve in endothelial cell,HUVEC)体外原代培养方法。通过倒置相差显微镜观察和比较,各组内皮细胞形态,用MTT法检测吸光度值,利用比色法测定细胞上清夜中的丙二醛(m alond ialdehyde,MDA)和一氧化氮(n itric oxide,NO)含量。结果MTT法结果显示:正常组、熊胆组与缺氧-再复氧组比较有显著差异(P<0.01)。缺氧-再复氧组的MDA含量增高,而NO含量降低,正常组和熊胆组中检测结果相反。结论熊胆冻干粉针剂对缺氧-再复氧损伤的血管内皮细胞有保护作用。

Abstract:ObjectiveTo study the protective effects of the lyophiled powder injection of bear bile on cultured vascular epithelial cells(VEC) induced by hypoxia-reoxygenation injury,and study the mechanisms of anti-atherosclerosis.MethodsBy using primary culture of human umbilical vein endothelial cells(HUVEC) in vitro, the VECs were divided into three groups.The morphological changes of VEC by the inverted microscope,the optical density(OD) value of cells were detected by MTT colorimetric method,content of MDA and NO in the supernatant fluid of cultured cells were detected by spectrophotometry.ResultsThe MTT assay showed that OD value had significant difference(P<0.01) between normal group,bear bile group and hypoxia-reoxygenation group.The MDA content increased and NO content decreased in the hypoxia-reoxygenation group,in contrast,MDA content decreased and NO content increased in the normal and bear bile groups.ConclusionThe results indicate that the lyophiled powder injection of bear bile has protective effects for the HUVEC injury induced by hypoxia-reoxygenation injury.
