雷公藤内酯醇对红白血病K562细胞株增殖和凋亡的影响

摘    要:目的探讨雷公藤内酯醇对人类红白血病K562细胞株增殖和凋亡的影响。方法运用四甲基偶氮唑盐(MTT)法、流式细胞术(FCM)等检测雷公藤内酯醇对K562细胞株增殖和凋亡的影响。结果雷公藤内酯醇能明显抑制K562细胞的增殖,呈时间与剂量依赖性,作用24h时其半数抑制浓度IC50为25nmol/L。且经雷公藤内酯醇处理后K562细胞G1/S期所占的百分比较对照组上升(P<0.05)。雷公藤内酯醇作用48h,其浓度的变化与K562细胞凋亡率具有相关性(r=0.97,P<0.05),而作用24h时,则无明显诱导凋亡的作用。结论雷公藤内酯醇有明确的抗白血病细胞增殖的能力,干扰K562细胞周期,上调细胞G/S期检测点,并具有促进细胞凋亡的作用。

Abstract:Objective To observe the effects of Triptolide on proliferation and apoptosis of erythroleukemia K562 in vitro.Methods The effects of Triptolide on proliferation and apoptosis of K562 cells were assayed by using cell culture, MTT and flow cytometry.Results Triptolide inhibited the proliferation of K562 lines in a dose- and time-dependent manner. IC_(50) of Triptolide was 25 nmol/L after treatment for 24 h. Triptolide could regulate the G_(1)/S transmit checkpoint. After treatment with Triptolide for 48 h, the apoptotic rate of K562 cells was correlated with the concentrations of Triptolide (r=0.97, P<0.05).Conclusion Triptolide possesses the potent of anti-leukemic cell proliferation, disturbs the cell cycle of K562, up-regulates the G_(1)/S transmit checkpoint and can enhance the apoptosis of K562 cells.
