雷藤氯内酯醇对大鼠生殖细胞的遗传效应

摘    要:雷公藤有效抗生育单体T_4(雷藤氯内酯醇)抗生育效果肯定,作用环节理想;对睾丸曲精细管细胞组合影响不大,主要作用于附睾,导致大量精子头尾分离。对精子头部形态没有明显影响,也不干扰精母细胞联会复合体配对及其功能,未见染色体易位的发生,也不引起精原细胞染色体畸变及微核发生率的增加,结果表明T_4对生殖细胞没有明显的遗传损伤效应。对T_4深入研究,可望发展成理想的男用避孕药。

Abstract:Clastogenicity of T_4 an effective contraceptive compound isolated from GTW in sperm head anomalies and synaptonemal complexes (Sc) of rat spermatocytes was analyzed in silver stained meiotic preparations. Adult male rats were given oral T_4(0.05mg/kg/d) for 7 weeks. Result indicated that the frequency of abnormal sperm heads in T_4-treated rats was similar with that of controls, T_4 did not affect synaptonemal complexes formation or function, as judged by the absence of pairing anomalies, Sc fragmentation, or presynaptic arrest. The unpaired lateral axes could be seen at zygotene, and at pachytene normal Sc could be observed. The behavior of the XY axes also appeared to be normal.
