高效液相色谱法测定犬血浆中磷酸苯丙哌林的质量浓度

目的建立用于测定犬血浆中磷酸苯丙哌林质量浓度的高效液相色谱法。方法以0.02 mol/L磷酸二氢钾(含0.5%三乙胺,磷酸调pH至4.0)-乙腈(60∶40)为流动相,尼卡地平为内标,血浆样品经正己烷-异丙醇的混合溶剂萃取后,采用高效液相色谱法测定血浆中磷酸苯丙哌林的质量浓度。结果磷酸苯丙哌林质量浓度在0.01～0.80μg/mL范围内与峰面积比线性关系良好(r2=0.998);平均相对回收率为96.89%～109.30%,日内和日间精密度RSD均小于12%,最低检测限为5 ng/mL。结论所用方法快速、准确、灵敏,可用于磷酸苯丙哌林的药代动力学研究。

Determination of Benproperine Phosphate in Dog Plasma by HPLC

Objective To establish an HPLC method to detect the mass concentration of benproperine phosphate in dog plasma.Methods The mobile phase consisted of 0.02 mol/L potassium dihydrogen phosphate solution（containing 0.5% triethylamine,adjusted pH to 4.0 with phosphoric acid）-acetonitrile（60：40） was used.Nicardipine was used as the internal standard.The plasma samples were pretreated by mixed solvent of N-hexane and avantin,then analyzed by HPLC.Results The linear range was 0.01-0.80 μg/mL（r2=0.998）.And the average recovery rate was 96.89%-109.30%.Both the within-day and between-day RSD were less than 12%.The minimum detective limit was 5 ng/mL.Conclusion This HPLC method is rapid,accurate,and sensitive,which can be applied to the pharmacokinetic study of benproperine phosphate.
