| 摘 要： | 建立了柱前衍生化-HPLC法同时测定三七中的18种游离氨基酸.以异硫氰酸苯酯为柱前衍生化试剂,采用C18色谱柱,以乙腈-0.05 mol/L乙酸钠缓冲液为流动相,梯度洗脱,检测波长254 nm.结果显示三七根中的游离氨基酸以精氨酸、丙氨酸和脯氨酸为主,约占18种游离氨基酸总量的80%.18种氨基酸的平均回收率为96.5%～103.4%,RSD均小于3%.

| Abstract: | A pre-column derivatization HPLC method was established for the determination of 18 free amino acids in Panax notoginsens.Phenylisothiocyanate was used as a derivatizing agent.A C_(18) column was used with the mobile phase of acetonitrile-0.05 mol/L sodium acetate buffer,at the detection wavelength of 254 nm.The results showed that the major components were arginme,alanine and proline,accounting for approximate 80%of free amino acids.The average recoveries were 96.5%-103.4%,with RSDs less than 3%. |
