阿尔兹海默病(AD)小鼠脑内P-糖蛋白(P-gp)的表达与功能分析

摘    要: 目的  研究P-糖蛋白(P-glycoprotein,P-gp)在APP/PS1双转基因阿尔兹海默病(Alzheimer′s disease,AD)模型小鼠脑中的表达与功能情况。方法   采用RT-PCR、real-time PCR和Western blot方法分析不同月龄的AD小鼠和野生型(wild type,WT)小鼠脑中P-gp表达的差异;采用尾静脉注射罗丹明123 (Rhodamine 123,Rh123),HPLC检测脑及血清中Rh123的含量,计算脑血分配系数,分析不同月龄的AD小鼠和WT小鼠脑中P-gp功能的差异。结果   3月龄时,AD小鼠脑内P-gp的表达与功能均明显低于WT小鼠;6月龄时,AD小鼠与WT小鼠无明显差异;9月龄时,AD小鼠脑内P-gp的表达与功能均明显高于WT小鼠;12月龄时,AD小鼠脑内P-gp的表达高于WT小鼠,功能却低于WT小鼠。结论  AD小鼠脑内P-gp的表达与功能与WT小鼠明显不同,并随月龄而变化。

Abstract:Objective  To study the expression and function of P-glycoprotein (P-gp) in the Alzheimer′s disease (AD) model mouse with APP/PS1 double transgene.Methods  RT-PCR,real-time PCR and Western blot were used to analyze the expression level of P-gp in the brain between the AD mice and wild type (WT) mice at different ages; Rhodamine 123 (Rh123) was injected by tail vein,and HPLC was used to detect the contents of Rh123 in brain and serum.The blood brain partitioning to was taken calculation analysis the function of P-gp between in the brain between AD mice and WT mice at different ages.Results   At the age of 3 month,the expression and function of P-gp in AD mice were significantly lower than that in WT mice; and at 6 month,there was no obvious difference between AD mice and WT mice; then at 9 month,the expression and function of P gp in AD mice were significantly higher than that in WT mice; but at 12 month,the expression of P-gp in AD mice was higher than that of WT mice,while the function was lower than that of WT mice.Conclusions   The expression and function of P-gp in AD mice is obviously different from that in WT mice,and the difference changed with ages.
