胰岛素对人肺腺癌细胞株A549影响的体外研究

目的：探讨用胰岛素提高人肺腺癌细胞A549生长代谢水平及其机制。方法：采用MTT比色法,分析胰岛素对肺腺癌A549细胞代谢的影响,用流式细胞仪作细胞周期分析,用Western Blot印迹方法检测细胞周期蛋白CyclinA和CyclinD1的表达情况。结果：胰岛素（2.0～32.0mU/mL）可促进人肺腺癌细胞A549的增殖（P〈0.05）,细胞周期结果显示胰岛素（8mU/mL）可使S期细胞增多。Western Blot印迹方法检测结果显示胰岛素（8mU/mL）后CyclinD1的表达明显增强,CyclinA的表达变化不明显。结论：胰岛素能促进人肺腺癌细胞株A549增殖,并且其增殖作用是可逆的。胰岛素主要作用于人肺腺癌细胞株A549细胞的S期。CyclinD1的表达增强在胰岛素促增殖机制中起着重要作用。

Effect of insulin on human adenocarcinoma cell strain A549 in vitro
