摘    要:目的研究外源性P物质(SP)对成纤维细胞迁移的影响及糖尿病小鼠创面愈合的作用。方法利用Transwell细胞迁移试验、划痕试验检测SP对小鼠成纤维细胞迁移能力的影响。筛选建模成功的糖尿病小鼠20只,随机分为对照组和SP组。分别于皮肤损伤后4、8、12、16、20 d,观察创面愈合情况并计算创面愈合率,并于7、14、21 d苏木精-伊红(HE)染色观察组织中炎症细胞聚集和肉芽组织形成情况,免疫组化检测Rac-1、JNK的表达水平。结果一定浓度SP可明显促进成纤维细胞迁移。损伤后第12天,SP组创面愈合率明显高于对照组(分别为(82.62±1.49)%、(57.24±1.3)%,差异有统计学意义(P<0.01);HE染色发现损伤后7 d,SP炎症细胞趋化明显;SP组创面Rac-1、JNK的阳性细胞(分别为28.3±2.5、36.2±1.6)均明显多于对照组(分别为11±3.2、24.4±3.2),差异有统计学意义(P<0.01)。结论SP可促进小鼠成纤维细胞迁移,促进创面愈合早期炎症细胞趋化和肉芽组织形成,从而加速创面愈合。

Abstract:Objective To investigate the effect of exogenous Substance P(SP)on fibroblast migration and wound healing in diabetic mice.Methods Transwell cell migration test and scratch test were used to detect the effect of SP on the migration ability of mouse fibroblasts. Twenty diabetic mice successfully modeled were screened and randomly divided into control group and SP group. The wound healing was observed and the wound healing rate was calculated at 4,8,12,16 and20 days after injury. The aggregation of inflammatory cells and granulation tissue formation were observed by H&E staining at 7,14 and 21 days. The levels of RAC-1 and JNK were detected by immunohistochemistry.ResultsA certain concentration of SP could significantly promote the migration of fibroblasts. On the 12 th day after injury,the wound healing rate of SP group was significantly higher than that of the control group(82.62±1.49)% vs.(57.24±1.3)%,which was statistically significant(P<0.001). H&E staining showed that SP inflammatory cells were chemotaxis significantly 7 days after injury,and granulation tissue formation increased at 14 days compared with the control group. The positive cells of RAC-1 and JNK were significantly more than those of the control group(28.3±2.5 vs 11±3.2,36.2±1.6 vs 24.4±3.2,respectively),showing statistically significant differences(P<0.01).Conclusion SP can promote the migration of fibroblasts in mice,promote the chemotaxis of inflammatory cells and the formation of granulation tissue in the early stage of wound healing,so as to accelerate wound healing.
