Doranidazole对结肠癌细胞的放射增敏作用

目的观察放射增敏剂-Doranidazole对结肠癌细胞的放射增敏作用。方法对5种人结肠癌细胞系施行2～50 Gy不同剂量放射线照射,照射后1～5 d检测细胞生长及死亡率,通过克隆形成实验绘制细胞存活曲线。从时间、剂量相关性、凋亡诱导等方面评价增敏效果。结果Doranidazole对照射后细胞的生长起到显著的抑制作用,其放射增感比为1.79;5种结肠癌细胞经30 Gy放射线照射后分别引起的18.7%、21.9%、16.7%、15.7%、19.5%的细胞死亡率,因Doranidazole的添加而增高至36.4%、33.8%、28.9%、22.2%、27.0%,并诱导了细胞的凋亡。这种增敏作用具有显著的时间和剂量相关性。结论Doranidazole对人结肠癌细胞系具有明确的放射增敏效果。

Doranidazole enhances the sensitivity of colorectal cancer cells to irradiation

Objective To investigate the radiosensitizing effects of Doranidazole on colorectal cancer cells.Methods The radiosensitizing effects of doranidazole were examined concerning the cell growth and cell killing of 5 human colorectal cancer cell hnes exposed to 2-50 Gy irradiation in vitro.The sensitizing effect of doranidazole on cell growth inhibition and cell killing was confirmed by time-course, dose-dependency,apoptotic analysis and microscopic observations.Results The cell proliferation evalu- ated by cell-clonogenic survival curves was significantly inhibited by 5 mM Doranidazole especially at the doses ranging from 10 Gy to 30 Gy of irradiation (The SER was 1.79).In 5 colorectal cancer cell lines, in which the percentage of dead cells was determined 4 days after 30 Gy irradiation in the presence of doranidazole,the percentage of dead cells was significantly increased from 18.7%,21.9%,16.7%, 15.7% and 19.5% to 36.4%,33.8%,28.9%,22.2% and 27.0% respectively.And the radiosensi- tizing effect of Doranidazole on colorectal cancer cells is time-and dose-dependent.Conclusion Doranidazole radiosensitized the cell killing and cell growth inhibition of colorectal cancer cells after high- dose irradiation.
