PITX2 shRNA对卵巢癌细胞H08910裸鼠移植瘤凋亡的影响

摘    要:目的探讨PITX2基因沉默对人卵巢癌细胞株H08910裸鼠移植瘤凋亡的影响。方法将重组PITX2 shRNA plasmid转染人卵巢癌细胞株H08910细胞,通过裸鼠移植瘤实验观察PITX2基因沉默对裸鼠移植瘤的生长抑制作用,同时,利用荧光定量聚合酶链反应(RT-PCR)和蛋白印迹法检测PITX2基因沉默对caspase-3、cytochrome-c和caspase-9表达的影响。结果与对照组(空载体组和未转染组)相比,PITX2 shRNA plasmid转染组人卵巢癌细胞株H08910细胞生长速度减慢,肿瘤体积缩小(P<0.05),而且caspase-3、cytochrome-c和caspase-9的表达明显升高(P<0.05)。结论 PITX2基因沉默可通过促进肿瘤细胞的凋亡而抑制裸鼠移植瘤生长。

Abstract:Objective To investigate the inhibitory effect of PITX2 gene silence on human ovarian cancer cell line H08910 tumor xenograft in nude mice.Methods Human ovarian cancer cell line H08910 was transfected with recombinant PITX2 shRNA plasmid,and the inhibitory effect of PITX2 gene silence on the tumor xenograft in nude mice was observed through the experiment of xenograft in nude mice.Meanwhile,the effects of PITX2 gene silence on the expressions of caspase-3,cytochrome-c and caspase-9 were detected by fluorescence quantitative PCR and Western blotting.Results Slower growth speed and smaller volume of human ovarian cancer cell line H08910 were found in PITX2 shRNA plasmid transfected group compared with control group (empty vector transfected group and untransfected group) (P〈0.05).And its expressions of caspase-3,cytochrome-c and caspase-9 significantly increased compared with control group (P〈0.05).Conclusion PITX2 gene silence could inhibit xenograft growth in nude mice through promoting the apoptosis of tumor cell.
