MSMHIV感染者特异性T细胞TCRQCDR3谱序初探

摘    要:目的监测HIV感染者的外周血样本TCRa链CDR3谱系变化,发现HIV特异性TCRTRAV家族。方法用本实验室已建立的“荧光定量PCR溶解曲线法监测HIV感染者TCRCDR3谱系漂移技术”,对22名健康男性、19名MSMHIV感染者的外周血样本TCRn链CDR3变化进行监测。结果MSMHIV感染者的TRAV4.2,TARVIO和TRAV23的单(寡)性表达频率要高于健康男性人群(P〈O.05),MSMHIV感染者的TRAV亚家族的缺失率(1.70%)虽然高于对照男性健康人群的TRAV亚家族的缺失率(0.80%),但二者没有统计学差异。结论在国内首次利用“荧光定量PCR溶解曲线法”监测HIV感染者TCRa链CDR3漂移变化,发现MSMHIV感染者的TCRAV家族存在单(寡)性增生和缺失现象,非正态分布现象增加,多样性减少。

Detection the TCR CDR3 shewing of TCRagene repertoire in the MSM HIV infected people

Abstract:Objective To monitor the change of the TCR ct CDR3 spectratyping and find the special T cell receptor (TCR) TRAV family in peripheral bloods of the HIV infections. Methods Monitor the change of the TCR ct CDR3 spectratyping in 19 MSM HIV infections and 22 healthy male populations by using the established method quantitative PCR fluorescence melting curve analysis in our laboratory. Results The TRAV4.2, TARV10 and TRAV23 monoclonal or oligoclonal in MSM HIV infections were higher than in healthy male people (P〈0.05), and the lack frequency of TRAV sub-family in MSM HIV infections was higher than in healthy male people, but no Significant statistics difference (P〈0.05) . Conclusion The phenomenon of monoclonal(or oligoelonal) proliferated, lacked, abnormal distribution increased and the diversity diminished in the TCR BV family were found in these of MSM HIV infections.
