端粒酶RNA与胃粘膜HpcagA及增殖细胞核抗原表达的关系

目的：探讨端粒酶RNA与胃粘膜幽门螺杆菌细胞毒素相关基因A（HpcagA）感染及增殖细胞核原（PCNA）表达的关系。方法取病理证实的各种胃粘膜病变的活检标本150例,应用原位PCR及免疫组化ABC法分别检测端粒酶RNA、HpcagA感染和PCNA表达。结果HpcagA感染各组的PCNA、端粒酶RNA阳性率随着胃粘膜病变的加重而上升（P＜0．05）。HpcagA感染组PCNA、端粒酶RNA阳性率均高于未感染组（P＜0．05）,其HpcagA核阳性与胃粘膜PCNSA及端粒酶RNA阳性表达呈显著正相关（r为0．747、0．671,P＜0．05）。结论HpcagA可通过刺激胃粘膜细胞过度增殖,使端粒酶RNA过度表达,而而启动及促进胃癌的发生发展过程。HpcagA核阳性可能是胃粘膜PCNA及端粒酶RNA阳性表达的机制之一。

Relation of telomerase RNA,HpcagA and expression of PCNA in gastric mucosa

Objectives To explore the expression of telomerase RNA of gastric mucosa in patients with Helicobacter pylori cytotoxin associated gene A (HpcagA) infection and Proliferating cell nuclear antigen (PCNA) expression. Methods The gastric biopsy specimens were taken from 150 patients.Telomerase RNA, HpcagA and PCNA were examined by in situ reverse transcription polymerase chain reaction and immunohistochemical staining respectivey. Results The expression of PCNA, telomerase RNA were instensive in HpcagA positive patients when compared with HpcagA negative patients( P＜0.05 ). The HpcagA nuclear signal was positively correlated with expression of PCNA and telomerase RNA. Conclusion HpcagA infection can enhance proliferative activities of gastric epithelia and overex-pression of telomerase RNA, which may promote the processes for gastric mucosa to become cancerous.
