抗原修复液pH值和抗体浓度对免疫组织化学技术检测PD-L2染色结果的影响

摘    要:目的:探讨不同浓度抗体和不同pH值的抗原修复液对免疫组织化学反应结果的影响,以提高免疫组织化学染色阳性率.方法:采用3种不同抗体浓度作预处理后,分别在pH值6.0和pH值9.0条件下,进行免疫组织化学SP法检测PD-L2分子在食管磷状细胞癌组织中的免疫组织化学染色效果.结果:PD-L2抗体稀释浓度1∶400的免疫组化染色效果优于抗体浓度1∶800;pH值9.0的抗原修复液处理后的免疫组化染色效果优于pH值6.0的抗原修复.结论:PD-L2抗体稀释浓度1∶400,pH值9.0抗原修复液处理后的免疫组化染色效果最佳,既可提高免疫组织化学反应阳性表达率,又能提高制片质量,是理想的检测PD-L2分子免疫组织化学反应的优化方案.

Antigen Retrieval Buffer PH Value and Antibody Concentration Effect on the Immunohistochemistry Staining of Detecting PD-L2
Abstract:Objective:To investigate the effects of different concentrations of antibodies and different pH values of the antigen retrieval buffer on results, and to improve the positive rate of immunohistochemistry.Methods:Pretreat samples in 3 different antibody concentration, respectively at pH6.0 and pH9.0,then using immunohistochemical staining results in the chemical SP(streptavidin-perosidase)method for detection of PD-L2 molecules in esophageal squamous cell carcinoma.Results:PD-L2 in antibody dilution 1∶400 immunohistochemical staining is better than that of 1∶800 group of antibody concentration;the pH9.0 after treatment with pH9.0 antigen retrieval buffer is better than pH6.0.Conclusion:PD-L2 antibody concentration 1∶400,immune treatment and pH9.0 antigen retrieval buffer is the optimized scheme,which can improve the positive reaction rate of immunohistochemical,but also improve the production quality,which is the ideal optimization scheme of PD-L2 molecular's immunohistochemistry.
