糖化终末产物对人结肠癌HCT116细胞中ChREBP表达的影响

摘    要:目的 研究糖化终末产物(advanced glycation end products, AGEs)对人结肠癌HCT116细胞中碳水化合物反应元件结合蛋白(carbohydrate responsive element binding protein, ChREBP)表达的影响。方法 采用RT-PCR检测AGEs处理的人结肠癌HCT116细胞中ChREBP及其下游基因的表达;使用N-乙酰半胱氨酸(NAC)干预氧化应激状态;运用Western印迹法观察在HCT116细胞中AGEs诱导对ChREBP表达的影响;运用MTS比色法观察AGEs诱导对HCT116细胞增殖的影响。结果 AGEs刺激后人结肠癌HCT116细胞中转录因子ChREBP表达增加(P<0.05);细胞经抗氧化剂NAC处理后,AGEs刺激所致ChREBP表达增加和HCT116细胞增殖均被明显抑制(P<0.05)。结论 AGEs通过激活氧化应激反应来上调人结肠癌HCT116细胞中的ChREBP表达。

Abstract:Objective To study the effects of advanced glycation end products (AGEs) on expression of carbohydrate responsive element binding protein (ChREBP) in human colorectal cancer HCT116 cells. Methods The expression of ChREBP mRNA in AGEs-treated human colorectal cancer HCT116 cells was detected with RT-PCR. After treatment with antioxidant N-Acetyl-L-cysteine(NAC), cell proliferation and ChREBP expression were examined with MTS assay and Western blot in AGEs-treated HCT116 cells, respectively. Results The expression of ChREBP mRNA was increased in AGE-treated HCT116 cells(P<0.05). ChREBP expression and cell proliferation were inhibited in AGE-treated HCT116 cells after NAC treatment(P<0.05). Conclusion AGEs up-regulate ChREBP expression in human colorectal cancer HCT116 cells through activation of oxidative stress.
