摘    要:目的 研究海洋真菌Aspergillus violaceus WZXY-m64-17中的抗菌活性成分。方法 利用硅胶、反相ODS、Sephadex LH-20、半制备HPLC等色谱学方法对菌株固体发酵物行分离纯化;利用NMR、MS、UV、IR等波谱学技术手段,结合文献数据比对,确定化合物的结构。并通过肉汤稀释法测定了单体化合物的抗金葡萄球菌和抗耐药菌活性。结果 分离鉴定了6个化合物,分别为2-hydroxyemodin(1)、emodin (2)、nidurufin (3)、versicolorin B (4)、sterigmatocystin (5)、terrequinone A(6),其中化合物4、6具有较好的抗菌活性。结论 海洋真菌Aspergillus violaceus能够产生一系列醌类化合物,其中部分化合物具有良好抗金葡萄球菌和抗耐药菌活性。

Antibacterial secondary metabolites from marine derived fungus Aspergillus violaceus WZXY-m64-17

Abstract:Objective To investigate the antibacterial chemical constituents of the marine-derived fungus Aspergillus violaceus WZXY-m64-17. Methods After larger-scale solid fermentation, the secondary metabolites were isolated from fermentation extracts based on column chromatography over silica gel, Sephadex-LH 20, C-18 ODS and semi-prepared HPLC. Their structures were elucidated through spectroscopic analysis of NMR, MS, UV and IR, as well as comparison with the data of literature. The antibacterial activities were assayed using broth dilution method. Results Six compounds were isolated from the EtOAc extract and their structures were identified as 2-hydroxyemodin (1), emodin (2), nidurufin (3), versicolorin B (4), sterigmatocystin (5), terrequinone A (6). Compounds 4 and 6 were found to inhibit the growth of Staphylococcus aureus and MRSA M6. Conclusion The fermentation extract of the fungus Aspergillus violaceus WZXY-m64-17 could produce secondary metabolites, and some of which showed antibacterial activity.
