乌司他丁对冷保存/再灌注胆管损伤的保护作用观察

目的：观察乌司他丁对冷保存/再灌注大鼠胆管损伤的保护作用。方法：雄性SD大鼠12只,随机分为两组,实验组肝脏保存液中加入乌司他丁,浓度为1000U/ml;对照组肝脏保存液中不加入乌司他丁,12h后电镜观察胆管上皮细胞情况,比较两组胆管上皮细胞凋亡指数、线粒体平均体积和数密度。结果：实验组大鼠电镜下可见胆管立方上皮细胞,线粒体无明显肿胀,绒毛形态较清晰,而对照组胆管上皮细胞电镜下观察,线粒体肿胀,基质均匀。胆管上皮细胞凋亡指数、线粒体平均体积和数密度两组大鼠间差异有统计学意义（P〈0.01或P〈0.05）,肝脏保存液中加入乌司他丁可以降低胆管上皮细胞的凋亡程度。结论：乌司他丁对冷保存/再灌注大鼠胆管上皮细胞有保护作用。

Protective effect of Ulinastatin on bile duct against cold preservation/reper-fusion injury

Objective: To observe the protective effect of Ulinastatin on bile duct against cold preservation/reperfusion injury. Methods: Twelve SD rats were randomly divided into two groups, experimental group and control group. Ulinastatin was added into liver preservative fluid in experimental group, while the control group didn't add, and the bile duct cellula epithelialis was observed with electron microscope 12 h later, and then the observation indexes including the apoptosis index, mitochondria mean volume and density of the bile duct cellula epithelialis were compared between the two groups. Results: Bile duct cuboidal epithelium could be observed under the electron microscope in rats of experimental group, and no obvious mitochondria swelling was observed, and the shape of the villi was quite clear; the mitochondria swelling, homogeneous matrix could be observed in the bile duct coboidal epithelium in rats of control group. The differences of apoptosis index, mitochondria mean volume and density showed statistical significances between two groups(P<0.01 or P<0.05). The addition of Ulinastatin into liver preservative fluid could decline the apoptosis degree of bile duct cuboidal epithelium. Conclusion: Ulinastatin added into liver preservative fluid can protect bile duct against cold preservation/reperfusion injury
