辅酶Q10对自然衰老小鼠皮肤超氧化歧化酶活性及病理学变化的影响

摘    要:目的 通过观察辅酶Q10对自然衰老小鼠皮肤超氧化歧化酶活性和组织病理学变化的影响,探讨辅酶Q10延缓衰老,减轻对皮肤细胞损伤的作用,为其研究开发提供实验依据.方法 将35只8月龄的SPF级昆明种雌性小鼠随机分为空白溶剂组、维生素E组、辅酶Q10低剂量组、辅酶Q10高剂量组、维生素E与辅酶Q10低剂量联合组,每组7只.采用经口灌胃法给药,1日1次,连续灌胃10周.实验终末用试剂盒法测定小鼠皮肤中的超氧化歧化酶(SOD)活性,光镜观察皮肤组织的病理学变化.结果 与空白溶剂组相比,辅酶Q10各剂量组的SOD活性不同程度升高,病理学观察可见辅酶Q10各剂量组小鼠皮肤表皮厚度增加,真皮纤维排列整齐.结论 辅酶Q10对自然衰老小鼠皮肤有保护作用,增加表皮厚度,改善真皮纤维结构,延缓皮肤的衰老.

Abstract:OBJECTIVE To study the SOD activity and pathological changes of the skin of intrinsic aging mice and discuss the effect of coenzyme Q10 on d elay aging and reduce skin cell damage,which can provide the experimental basis for the research and development of coenzyme Q10.METHODS 35 e ight months old female mice of SPF were randomly divided into blank group,vita mi n E group,low dose group of coenzyme Q10,coenzyme Q10 group and vitamin E comb ined coenzyme Q10 in low dose group,7 mice in each group.The samples was intra gastrically administrated to the mice,once a day,for 10 weeks.The activity of superoxide dismutase (SOD) in mouse skin was measured by Reagent kit method.Th e pathological changes of skin were observed by light microscope.RESULT S Compared with the blank group,the SOD activity of coenzyme Q10 in ea ch dose group has raised than that in the blank group.The results of light micr oscope observation showed that coenzyme Q10 in each dose group increase the epid ermal thickness of skin and the dermal fiber was compact and neat.CONCL USION Coenzyme Q10 has a protective effect on the skin of natural aging mice and delay skin aging by increasing the thickness of epidermis,improving t he structure of dermal fiber.
