宜宾地区健康新生儿淋巴细胞免疫分型研究

摘    要:目的 研究宜宾地区健康新生儿的淋巴细胞免疫分型特点,探讨健康新生儿与正常成人之间淋巴细胞免疫分型的差异.方法 选取2015年1月至2016年12月在宜宾市第二人民医院分娩及儿童保健科正常体检的新生儿26例作为新生儿组,随机选取同期本院健康体检者44例作为成人组.采集新生儿足跟血60μL,成年人全血2~3 mL,所有标本均在24 h内用BD FACSCanto II流式细胞仪完成检测.结果 与成人相比,宜宾地区新生儿末梢血CD3+T淋巴细胞百分比(81.40±6.04)%vs(70.10±5.83)%]、CD3+CD4+CD8-T淋巴细胞百分比(57.50±8.38)%vs(38.90±4.83)%]及CD3+T淋巴细胞绝对计数(3407.50±945.57)×106个/μL vs(1429.50±376.44)×106个/μL]、CD3+CD4+CD8-T淋巴细胞绝对计数(2396.00±671.27)×106个/μL vs(791.70±237.18)×106个/μL]、CD3-CD19+B淋巴细胞绝对计数(407.60±281.40)×106个/μL vs(249.50±92.46)×106个/μL]、CD3+CD4-CD8+T淋巴细胞绝对计数(980.40±396.68)×106个/μL vs(531.70±155.51)×106个/μL]显著增高,差异均有统计学意义(P<0.05),而CD3+CD4-CD8+T淋巴细胞百分比(23.20±6.64)%vs(26.20±4.27)%]、CD3-CD16+CD56+NK细胞百分比(6.00±2.91)%vs(16.50±5.64)%]及CD3-CD16+CD56+NK细胞绝对计数(252.90±151.38)×106个/μL vs(325.80±129.25)×106个/μL]显著降低,差异也均有统计学意义(P<0.05);新生儿末梢血CD3-CD19+B淋巴细胞百分比与成人组之间比较差异无统计学意义(P>0.05).结论 宜宾地区健康新生儿末梢血淋巴细胞免疫分型与正常成年人之间存在差异,呈现了淋巴细胞随着人类的生长发育而表现出的不同特点.

Abstract:Objective To investigate the characteristics of peripheral blood lymphocyte subsets in healthy new-borns in Yibin regions, and to explore the lymphocyte subsets differences between healthy newborns and normal adults. Methods From January 2015 to December 2016, 26 newborns delivered in the delivery room or had a physical exami-nation in Department of Child Care in the Second People's Hospital of Yibin were taken as the newborn group, and 44 normal adults of physical examination were randomly selected as the adult group. A total of 26 heel blood sample from healthy newborns and 44 whole blood samples from normal adults were collected. The percentages and absolute counts of lymphocyte subsets were detected by BD FACSCanto II flow cytometry within 24 hours. Results The percentages of CD3+T lymphocyte, CD3+CD4+CD8-T lymphocyte, and absolute counts of CD3+T lymphocyte, CD3+CD4+CD8-T lymphocyte, CD3+CD4-CD8+T lymphocyte, B lymphocyte in the newborn group were (81.40±6.04)%, (57.50±8.38)%, (3407.50±945.57)×106/μL, (2396.00±671.27)×106/μL, (980.40±396.68)×106/μL, (407.60±281.40)×106/μL, respectively, which were significantly higher than corresponding (70.10±5.83)%, (38.90±4.83)%, (1429.50±376.44)×106/μL, (791.70± 237.18)×106/μL, (531.70±155.51)×106/μL, (249.50±92.46)×106/μL in the adult group (P<0.05), while the percentages of NK cell, CD3+CD4-CD8+T lymphocyte and absolute counts of NK cell were (6.00 ± 2.91)%, (23.20 ± 6.64)%, (252.90 ± 151.38) × 106/μL, respectively, which were significantly lower than corresponding (16.50 ± 5.64)%, (26.20 ± 4.27)%, (325.80 ± 129.25) × 106/μL in the adult group (P<0.05). There was no significant difference in the percentages of CD3-CD19+B lymphocyte between the newborn group and adult group (P>0.05). Conclusion There is a difference be-tween healthy newborns and normal adults in the immunocytochemistry of peripheral blood lymphocytes in Yibin re-gions, which shows the different characteristics of lymphocytes with human growth and development.
