紫杉醇水蛭素复合物对兔血管平滑肌细胞和内皮细胞增殖与迁移的影响

摘    要:目的观察不同浓度紫杉醇水蛭素复合物对兔血管平滑肌细胞(smooth musele cell,SMC)和内皮细胞(endothelial cell,EC)增生、迁移的影响。方法培养兔血管SMC和EC,建立细胞共培养体系,模拟紫杉醇水蛭素复合物对血管SMC及EC的作用方式,观察不同浓度紫杉醇水蛭素复合物对兔血管SMC及EC的DNA合成、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)蛋白表达的影响,测定不同浓度紫杉醇水蛭素复合物对血管SMC及EC的迁移率。结果溶剂对照组SMC及EC的氚-胸腺嘧啶脱氧核苷(^3H-TdR)掺入、PCNA蛋白表达、迁移与空白对照组相比差异均无显著性。大、中、小剂量紫杉醇水蛭素复合物呈浓度依赖地抑制SMC的^3H—TdR掺入、PCNA蛋白表达和迁移(n=6,P〈0.05);在大、中剂量之间,紫杉醇水蛭素复合物呈浓度依赖地抑制EC的^3H—TdR掺入、PCNA蛋白表达和迁移(n=6,P〈0.05);但在小剂量组紫杉醇水蛭素复合物对EC的^3H—TdR掺入、PCNA蛋白表达和迁移有抑制倾向,但与空白对照组相比差异均无显著性。结论小剂量的紫杉醇水蛭素复合物可显著抑制兔血管SMC的增生与迁移,但抑制EC的增生与迁移不明显。

Effects of paclitaxel hirudin complex on proliferation and migration of vascular smooth muscle cells and endothelial cells of rabbits

Abstract:Objective To observe the effects of paclitaxel hirudin complex in different concentrations on the proliferation and migration of vascular smooth muscle cells ( SMC ) and endothelial cells ( EC ) in rabbits. Methods The rabbit 's SMC and EC were cultured and a cocuhure system was established. The effect model of paclitaxel hirudin complex on SMC and EC was simulated. The influences of paclitaxel hirudin complex in different concentrations were observed on DNA synthesis and protein expression of proliferating cell nuclear antigen (PCNA) of SMC and EC. The migration rates of paclitaxel hirudin complex to SMC and EC were detected. Results There was no significant difference in ^3 H-TdR incorporation and PCNA expression and migration of SMC and EC between the solvent control group and control group. Paclitaxel hirudin complex in high, medium and low concentrations inhibited ^3 H-TdR incorporation and PCNA expression and migration of SMC in a concentration-dependent manner( n = 6,P 〈 0.05 ), while it in high and medium concentrations inhibited 3 H-TdR incorporation and PCNA expression and migration of EC in a concentration-dependent manner( n = 6, P 〈 0.05 ). Although paclitaxel hirudin complex in low concentration had an inhibitive trend to 3 H-TdR incorporation and PCNA expression and migration of EC, the difference was not significant compared with the control group. Conclusion Paclitaxel hirudin complex in low dose can inhibit significantly the proliferation and migration of SMC in rabbits but has no obvious inhibitive effect on the proliferation and migration of EC.
