乳腺癌患者外周血、淋巴结、癌组织中免疫细胞构成分析

摘    要:目的分析乳腺癌患者外周血、淋巴结、癌组织免疫细胞构成。方法20例健康志愿者及40例乳腺癌患者外周血、18例乳腺癌患者反应性增生淋巴结及40例乳腺癌肿瘤组织细胞悬液用于流式细胞术检测,分析其单个核细胞群中CD3+、CD4+、CD8+、CD19+、CD56+细胞百分比及CD34-Lin- HLA-DR+细胞群中细胞CD11c+ CD123- (pDC1)、CD11c-CD123+( pDC2)百分比,计算 CD4/CD8比值以及DC1/DC2比值。结果乳腺癌患者外周血CD3+、CD4+、CD8+、CD19+、CD56+细胞百分比、CD4/CD8比值与对照组比较,差异均无统计学意义(P>0.05),pDC1/pDC2比值显著低于对照组(P<0.05);反应性增生淋巴结组织亚群构成为:CD3+(54.5±6.8)%、CD4+(37.8±4.9)%、CD8+(13.1±5.5)%、CD19+(5.9±3.4)%、CD56+(1.9±0.4)%、CD4/CD8为(3.6±1.3),DC1/DC2为(3.6±1.3)%。无法取得正常组织中的值,未作对照分析。瘤组织中CD3+(62.4±17.5)%、CD4+ (32.0±8.2)%、CD8+(26.9±10.5)%、CD4/CD8(1.3±0.4)、CD19+(2.2±2.0)%、CD56+(2.3±1.6)%,DC数不足以分析。结论乳腺癌患者外周血、淋巴结、肿瘤组织中免疫细胞构成,与对照组比较表现为抗肿瘤特征,循环中pDC1/pDC2比值减低可能与DC1在引流淋巴结中分布增加有关。

Abstract:Objective To explore the components of immunocytes in peripheral blood,lymph gland and cancer tissue of patients with breast cancer.Methods The peripheral blood from 20 healthy volunteers and 40 cases of breast cancer,and cell supension from 18 cases of reactive hyperplasia lymph gland tissue and 40 cases of tumor tissues from patients with breast cancer were collected for detection of the percent of CD_3~+、CD_4~+、CD_4~+、CD_(19)~+、CD_(56)~+ cells in their mononuclear cells and the percent of CD_(110)~+ CD_(123)~-(DC_1),CD_(110)~-CD_(123)~+(DC_2)cells in their CD_(34)~-Lin~-HLA-DR~+ cells by flow cytometry.The ratio of CD_4/CD_8 and DC_1/DC_2 was analyzed.Results There was no significant difference between the patients with breast cancer and controls in percent of CD_3~+,CD_4~+,CD_8~+,CD_(19)~+,CD_(56)~+ cells and the ratio of CD_4/CD_8 in the peripheral blood (P>0.05).But the ratio of DC_1/DC_2 in the peripheral blood of patients was markedly lower than that in the controls(P<0.05).The percentage of CD_3~+,CD_4~+,CD_(56)~+,CD_(19)~+ and CD_8~+ cells was (54.5±6.8)%,(37.8±4.9)%,(13.1±5.5)%,(5.9±3.4)%,(1.9±0.4)% and the ratio of CD_4/CD_8 and DC_1 / DC_2 was 3.6±1.3 and 3.6±1.3 separately in the tissues of reactive hyperplasia lymph glands.The percentage of CD_3~+,CD_4~+,CD_(56)~+,CD_(19)~+ and CD_8~+ cells was separately (62.4±17.5)%,(32.0±8.2)%,(26.9±10.5)%,(2.2±2.1))%,(2.3±1.6)%,the ratio of CD_4/CD_8 was 1.3±0.4 and the number of DCs were too less to analyse in the tissue of cancer.Conclusion The components of immunocytes in peripheral blood,lymph gland and cancer tissue of patients with breast cancer showed antitumor character,and the decrease in the ratio of DC_1 / DC_2 in the circulation may be related to the increase in the number of DC_1 distributing in the drainage lymph gland.
