Abstract:Objective: To investigate the mechanism of intense noise-induced cochlea cells death in guinea pig and the role of CHOP/Gadd153 during the procedure of cochlea cells apoptosis.Methods: Thirty-two guinea pigs were randomly divided into four groups.The guinea pigs in the experiment groups were exposed to 4 kHz narrow band noise at 120 dB SPL for 4 h.After the noise expose of 1,4 or 14 days on the experiment guinea pigs,auditory brainstem response(ABR) of the guinea pigs in experiment and control groups were tested before killed.Apoptosis was tested by terminal deoxynucleotidyl Transferase(TdT)-mediated deoxyuridine triphosphate(d-UTP) nick and labeling method(TUNEL).Expression of CHOP/Gadd153,Bcl-2 were tested by immunohistochemistry and Western blot methods.Results: There were higher expressions of TUNEL-positve cells in the organ of Corti,stria vascularis,and spiral ganglion cells of experiment groups and significant differences were observed as compared with the control group(P<0.01).CHOP/Gadd153 expression was higher in 1-day,4-day and 14-day groups than that of the control(P<0.01),however,there was no statistically difference(P>0.05) among 1-day,4-day and 14-day groups.In the experimental groups,Bcl-2 was negatively expressed while it had significantly high expression(P<0.01) in the control group.Conclusion: Intense noise can lead to the apoptosis of guinea pig cochlea cells through increasing CHOP/Gadd153 protein expression and decreasing expression of Bcl-2 protein.
