幽门螺杆菌基因型与慢性胃炎和消化性溃疡的相关性

摘    要:目的 探讨Hp基因型与慢性胃炎、消化性溃疡的关系。方法 对标准株和临床分离株幽门螺杆菌空泡毒素基因vacA和致病岛基因的cagA、cagE、cagT片段进行PCR扩增,对基因组DNA进行随机扩增多态性分析(RAPD),并以Hela细胞测定全部菌株的VacA毒素活性。SPSS13.0软件聚类分析进行基因分型。结果 全部菌株被分为3个型别,慢性胃炎、消化性溃疡和非胃炎非溃疡分别有自己的优势基因型,且各型之间的总体分布差异具有统计学意义。结论 Hp基因型与疾病类型有关,空泡毒素基因和致病岛基因可作为Hp基因分型的靶基因,RAPD是有效的基因分型手段。

Abstract:Objective To explore relationship between genotypes of Helicobacter pylori and chronic gastritis and peptic ulcer. Methods The vacA and cagA,cagE,cagT genetic fragments of standardized strains and clinically isolated Helicobacter pylori strains were amplified by PCR and the diversity of genome was analyzed by random amplified polymorphic DNA(RAPD) technology.The expression of VacA of all strains were detected with Hela cells cultured with Helicobacter pylori extract. Results All Helicobacter pylori strains were classified into 3 genotypes.Each of chronic gastritis or peptic ulcer or no-gastritis and no-ulcer has its own dominant genotype.There significant differences were observed in the distribution of genotypes. Conclusion The genotype of Helicobacter pylori is associated with the kind of illness and vacA,cagA,cagE,cagT can be taken as ideal indicators for gene typing of Helicobacter pylori.While RAPD was an effective method for analysis of genome DNA diversity.
