人胚胎生殖干细胞的体外培养研究

本实验在综合分析目前国内外人胚胎生殖细胞（EG细胞）的培养条件后，采用组织块培养法，以同源胚胎的成纤维细胞为饲养层，在不添加任何细胞因子的情况下，对人EG细胞进行体外培养。该培养体系确保了在培养过程中人EG细胞有充足的来源，避免了胰酶以及一些机械操作对原始生殖细胞（PGCs）的损伤，同时也避免了由于用小鼠成纤维细胞做饲养层，而对培养体系造成的异种蛋白的污染。这是关于建立人EG细胞合适的体外培养体系的一次有意义的探索。

Research on cultured in vitro of human embryonic germ cells

This experiment after the generalized analysis the domestic bystander EG cell raise condition, uses the organization block raise law at present, becomes the ciliary cell take the homologous embryo as the raising level, and does not increase any cell factor in the situation, carries on in vitro raise to the human EG cell. The cultured system guaranteed the human EG cell had the sufficient origin in the raise process, has avoided the pancreas enzyme as well as some mechanical operations to the PGCs damage, simultaneously avoided because became the ciliary cell with the mouse to make the raising level, but to raised the heterogeneous protein pollution which the system created. This is about establishes the human EG cell appropriate in vitro raise system one time to have the significance exploration.
