半夏与其伪品掌叶半夏的RAPD鉴别

摘    要:目的建立半夏及其伪品掌叶半夏的快速简单的分子鉴定方法。方法 SDS法提取半夏和掌叶半夏的基因组DNA,20个10碱基随机引物(S1-S20,上海生工)用于RAPD-PCR分析。结果 20个引物中有7个有效稳定的引物在半夏和掌叶半夏之间能显示出多态性指纹图谱,其中S10和S17在半夏和掌叶半夏之间显示较大差异。结论利用引物S10和S17进行RAPD-PCR能有效鉴别半夏和掌叶半夏。

Abstract:Obiecfive To establish a rapid and simple molecular identification method for Pinellia ternata and Pinellia pedatisecta. Methods Genomic DNA was isolated from the dried tubers of P ternata and P pedatisecta by modified SDS method. Twenty 10-mer random primers (S1-S20) were used for RAPD analysis. Results Seven efficient and stable primers, showing distinct and polymorphic fingerprints, were selected from the 20 primers. The primers S 10 and S 17 were selected to differentiate Pinellia ternata from Pinellia pedatisecta. Conclusion RAPD analysis using the primer S10 and S17 would be accurate and reliable to identify P. ternata from P pedatisecta.
