射频消融治疗前后大鼠树突状细胞的变化

摘    要:目的探讨肝脏射频消融(RFA)对正常大鼠树突状细胞(DC)的影响。方法26只健康SD大鼠随机分为治疗7d组(9只)、治疗14d组(9只)和对照组(8只),对照组不做治疗直接处死取材,后两组分别在RFA治疗后7d和14d处死取材。取外周血、消融区周边0.5cm内肝组织(对照组取正常肝组织)及脾脏,采用流式细胞术检测树突状细胞表面OX62、OX6、CD86表达水平。结果对照组大鼠OX62 细胞占外周血单个核细胞比率为(0.37±0.11)%,治疗7d组增高为(0.59±0.23)%,两组相比差异有显著性(P<0.05)。对照组肝脏、脾脏OX62 细胞占单个核细胞比率分别为(17.50±9.51)%、(10.36±3.21)%,射频消融后分别增高至(34.94±15.14)%、(18.03±5.70)%(P<0.05);射频后肝组织局部和脾脏DC表面OX6阳性表达率明显增高,与对照组大鼠相比差异有显著性(P<0.05)。结论射频消融后大鼠外周血DC前体细胞数量明显增多,肝脏局部和脾脏DC数量增多,OX6 表达率增高。射频治疗后机体DC的数量及功能发生变化,为进一步研究射频治疗肝肿瘤对机体抗原递呈功能的影响奠定了基础。

Abstract:Objective To investigate the effect of radiofrequency ablation (RFA) on dendritic cells (DCs) in healthy rats. Methods Twenty-six healthy SD rats were divided into RFA 7 d group (n=9), RFA 14 d group (n=9) and control group (n=8). The rats in control group were killed without treatment and the rats in RFA 7 d and 14 d groups were killed on the 7th day and 14th day after RFA respectively. Peripheral blood, liver tissue around the ablation region and spleen were taken out. The expressions of OX62, OX6 and CD86 of DCs were analyzed with flowcytometry. Results OX62+ cells accounted for (0.37±0.11)% of mononuclear cells in peripheral blood in normal rats and increased to (0.59±0.23)% in RFA 7 d group. There were significant differences between the two groups (P<0.05). OX62+ cells accounted for (17.50±9.51)% and (10.36±3.21)% of mononuclear cells in liver and spleen respectively. After RFA, the ratio increased to (34.94±15.14)% and (18.03±5.70)% respectively(P<0.05). The expressions of OX6 on DCs increased significantly in liver and spleen after RFA. There were significant differences compared with control group (P<0.05). Conclusion The precursor DCs in peripheral blood and DCs in liver and spleen increase significantly after RFA. The expressions of OX6 on DCs in liver and spleen increased after RFA. The change of number and function of DCs in normal rats after RFA may contributes to further study of the effect of RFA on DCs in body with hepatic carcinoma.
