喉癌组织HMGA2表达及其临床意义的探讨

目的:探讨喉癌组织中HMGA2的作用及其临床意义.方法:采用实时荧光定量PCR (RQ-PCR)检测HM-GA2基因在20例喉癌组织和癌旁正常黏膜上皮组织中mRNA的水平,同时制作组织芯片应用免疫组化检测其在50例喉癌组织和癌旁正常黏膜上皮组织中蛋白水平的表达.结果:喉癌组织和癌旁正常黏膜上皮组织HMGA2 mRNA的相对表达量分别为0.358±0.261和0.203±0.151,HMGA2蛋白的阳性率分别为86.0％ (43/50)和27.3％(12/44),两组比较差异有统计学意义,P值均＜0.05；HMGA2的表达与与患者性别、年龄无显著相关性,P＞0.05；与TNM分期和淋巴结转移有显著相关性,P＜0.05.结论:HMGA2在喉癌组织中的表达与肿瘤的发生、侵袭和转移密切相关,可能成为喉癌新的肿瘤标志或预后因子.

Expressions of HMGA2 and their clinical significance in laryngeal squamous cell carcinoma

OBJECTIVE: To investigate the expression of HMGA2 in laryngeal squamous cell carcinoma(LSCC) and their relationship with clinicopathological features.METHODS:The mRNA expression of HMGA2 was determined by real-time quantitative PCR in 20 laryngeal carcinoma tissues and para-carcinoma normal tissues.Subsequently,high-quality tissue microarray(TMA) was made with fifty matched pairs of the tumor samples including fifty all stages of LSCC and fifty para-carcinoma normal tissues.Immunohistochemistry was used to quantitatively analyze the protein levels in TMA.RESULTS:The mRNA expression of HMGA2 in LSCC,the para-carcinoma normal tissues were 0.358±0.261,0.203±0.151,and the positive expression rate of HMGA2 protein were 86.0%(43/50) and 27.3%(12/44),comparison between the two groups showed no significant difference(P<0.05).There were no significant correlation between the copies of HMGA2 and gender,age(P>0.05),but related to TNM stage and lymph node metastases,significantly(P<0.05).CONCLUSIONS:Based on these findings,overexpression of HMGA2 in LSCC suggests they may be participating in the genesis and development of LSCC.HMGA2 can be useful tumor markers and prognostic factors for LSCC.
