雷公藤内酯对海入酸致痫大鼠神经元caspase3和caspase9蛋白表达的影响

摘    要:目的:通过检测海马神经元caspase3和caspase9蛋白表达,探讨雷公藤内酯抑制神经元凋亡的机制。方法:结晶紫染色观察海马神经元形态,免疫组化法检测海马神经元caspase3和caspase9蛋白表达水平。结果:结晶紫染色结果显示,海人酸组中,海马神经元变性或丢失,胞体皱缩,形态不规则。雷公藤内酯干预组中海马神经元的数量和形态与对照组相似,胞浆清晰淡染,核仁清楚。免疫组化染色显示,与对照组比较,海人酸组海马神经元caspase3和caspase9蛋白表达水平显著增高(P〈0.05);与海人酸组相比,雷公藤内酯干预组神经元caspase3和caspase9蛋白表达水平显著减少(P〈O.05)。结论:雷公藤内酯可下调海人酸致痫大鼠海马神经元caspase3和caspase9蛋白表达.从而抑制神经元凋亡。

Abstract:Objective : To investigate the mechanism of triptolide inhibiting neurons apoptosis through detecting the expressions of caspase3 and caspase9 in the hippocampal region. Methods : Crystal Violet Staining was used to observe the morphological change of the hippoeampal neuron. Immunohistochemistry staining was used to examine the expressions of caspase3 and caspase9 in the hippocampal region. Results : Crystal Violet Staining showed that the neurons of the hippocampal region exhibited irregular morphology and body shrinked and numbers decreased partly in kainite group. However, the neurons of triptolide treatment group showed clear cytoplasm dyed and nucleoli, and had the similar number and morphology compared with the control group, hnmunohistochemistry staining showed that the expressions of easpase3 and caspase9 on neurons in hippocampal region in kainite group were increased significantly than that in control group ( P〈0.05 ) .Compared with kainite group, the expressions of caspase3 and easpase9 on neurons in hippoeampal region in triptolide treatment group were obviously decreased (P〈0.05). Conclusion: Triptolide can inhibit the expressions of caspase3 and caspase9 in the hippocampal neuron, and finally inhibit neurons apoptosis.
