黄芪对实验性自身免疫性脑脊髓膜炎小鼠的治疗作用及机制研究

摘    要:目的:观察中药黄芪对实验性自身免疫性脑脊髓膜炎(EAE)小鼠的治疗作用,并探讨其免疫调节机制。方法:建立C57BL/6小鼠EAE模型,通过5级临床症状评分观察黄芪对EAE小鼠的治疗作用;HE染色观察黄芪对EAE小鼠脊髓炎症细胞浸润的影响;CCK-8法检测脾脏髓鞘少突胶质细胞糖蛋白(MOG)多肽MOG35-55特异性T细胞增殖情况;ELISA法检测脾脏MOG35-55特异性T细胞培养上清液中IFN-γ、TNF—α、IL-17、IL-4的水平。结果:黄芪能够有效抑制EAE小鼠的临床症状,减轻中枢神经系统的炎性反应,抑制MOG35-55特异性T细胞增殖及IFN-γ、TNF-α、IL-17的分泌,促进IL-4的分泌。结论:黄芪能够有效抑制中枢神系统的自身免疫反应,这种作用与其抑制MOG35-55特异性T细胞增殖,减少Th1、Th17型细胞因子分泌,促进Th2型细胞因子分泌相关。

Abstract:Objective : To observe the therapeutic effects of Radix Astragali seu Hedysari on experimental autoimmune encephalomyelitis (EAE) mouse, and to explore its immunomodulatory mechanisms. Methods : The C57BL / 6 mouse models were established. The therapeutic effects of Radix Astragali seu Hedysari on EAE mouse were observed by clinical symptom scores ; the inflammatory infiltration of spinal cord tissue was examined by HE staining ; the proliferation of MOG35.55 -specific T-lymphocyte was measured by the CCK-8 method; the ELISA assay was used to detect the production of cytokines including IFN-γ、TNF-α, IL-17 and IL-4 in cell culture supematants. Results : Radix Astragali seu Hedysari can effectively inhibit the clinical symptoms of EAE in mouse,reduce the inflammatory response of central nervous system, inhibit the proliferation of MOG35-55-specific T-Iymphocyte, decrease the secretion of IFN-γ、TNF-α and IL-17,and promote the secretion of IL-4. Conclusion : Radix Astragali seu Hedysari can effectively inhibit the autoimmune reactions in the central nervous system ; this effect is related to its inhibition of MOG35-55 -specific T cell proliferation, reduction of Th1 and Th17 cytokine secretion, and promotion of Th2 cytokines secretion.
