针刺对铝中毒SD痴呆大鼠脑组织内细胞凋亡及Bcl-2影响的实验研究

目的:从细胞凋亡及凋亡相关基因的蛋白表达方面探讨针刺治疗老年性痴呆(Alzheimer’s disease,AD)的作用机理。方法:采用AlCl3中毒法制成SD大鼠老年性痴呆模型,连续针刺治疗20d。通过空白组、模型组、西药组及针刺组之间的对照,观察针刺该组穴位对大鼠"Y"型电迷宫行为学改变,应用TUNLE法和免疫组化法对所设各组SD大鼠脑组织细胞凋亡的数目及Bcl-2免疫阳性细胞进行观察。结果:治疗后,Bcl-2免疫阳性细胞数西药组、针刺组与模型组有显著性差异(均P0.05),西药组、针刺组多于模型组;细胞凋亡数目西药组、针刺组与模型组有显著性差异(均P0.05),西药组、针刺组少于模型组。结论:铝中毒法复制AD大鼠可行。针刺可减少老年性痴呆大鼠DNA双键断裂(TUNLE阳性),提高海马皮层Bcl-2的含量。

Experimental Study on Effect of Acupuncture on Brain Apoptosis and Bcl-2 in Aluminum-poisoned SD Rats

Objective： To explore the mechanism of acupuncture treatment of senile dementia （ Alzheimer’s disease,AD） in terms of protein expression of apoptosis and apoptosis-related gene e. Methods： SD rats were made with AlCl3 poisoning method to have a continuous acupuncture treatment for 20 d. The blank group,model group,Western medicine group and the acupuncture group were compared to observe＂Y＂-type electric maze behavioral changes of the acupuncture group after the treatment. TUNLE method and immunohistochemistry were applied to observe the number of SD rat brain apoptosis and Bcl-2 immunopositive cells of different groups. Results： After the treatment,the number of Bcl-2 positive cells in the immune medicine group,acupuncture group and model group were significantly different （ all P 0. 05） ,the number of Bcl-2 positive cells in the Western medicine group and acupuncture group is more than that of the model group; the number of apoptotic cells in the Western medicine group, acupuncture group and model group were significantly different （ P 0. 05） ,the number of apoptotic cells in the Western medicine group and acupuncture group is less than that of the model group. Conclusion： It is feasible to replicate AD rats with the aluminum intoxication method. Acupuncture can reduce DNA double bond fracture of senile dementia rat （ TUNLE positive） ,and increase Bcl-2 content of hippocampal cortex.
