hTERC基因在不同级别宫颈病变中的表达与意义

摘    要:目的探讨hTERC基因扩增在不同级别宫颈病变中的表达及其临床意义。方法应用荧光原位杂交技术(FISH)检测150例宫颈脱落细胞中hTERC基因的表达情况,其中106例为高危型HPV阳性,44例为阴性。结果 hTERC基因扩增在组织学正常或炎症、CINⅠ、CINⅡ、CINⅢ及浸润性宫颈癌中的表达率分别为5.0%、22.2%、57.1%、81.3%、96.3%,各组间hTERC基因异常扩增率差异均有统计学意义(P〈0.05);HPV阳性患者中hTERC基因扩增率为72.6%(77/106),阴性患者中hTERC基因扩增率为9.1%(4/44);hTERC基因扩增的患者中,HPV阳性所占的比例为95.1%(77/81),阴性所占比例仅为4.9%(4/81),两者比较差异有统计学意义(P〈0.05)。结论高危型HPV感染可能是hTERC基因异常扩增的早期事件,hTERC基因异常扩增检测可能成为宫颈癌筛查及宫颈病变鉴别诊断的有效方法

Expression and significance of human telomerase gene in cervical cancer and cervical intraepithelial neoplasia

Abstract:Objective To evaluate the expression and the clinical significance of genomic amplification of the human telomerase gene(hTERC) in cervical intraepithelial neoplasia(CIN).Methods The hTERC gene was detected by fluorescence in situ hybridization(FISH) using 150 cytologic samples of cervix,including 106 cases with high-risk HPV infection and 44 without.Results Amplification of hTERC gene in the normal or inflammation samples,CIN,CINⅡ,CINⅢ and cervical cancer were 5.0%,22.2%,57.1%,81.3% and 96.3%,respectively,it showed significant differences among the five groups(P<0.05).Amplification of hTERC gene in 106 cases with HPV infected was significant more than 44 cases without HPV infection,72.6% vs 9.1%.In the cases with amplification of hTERC gene,cases with HPV infected was significant more than cases without HPV infection,95.1% vs 4.9%(P<0.05).Conclusion High-risk HPV infection may be an early stage event which related to amplification of hTERC gene,and detectation of amplification of hTERC gene may be an effective method to screen cervical cancer and CIN.
