“劳倦过度、房室不节”肾阳虚模型小鼠睾丸乳酸脱氢酶-X活性的测定

摘    要:[目的]观察"劳倦过度、房室不节"肾阳虚小鼠睾丸乳酸脱氢酶(LDH)总活性及精子特异酶LDH-X活性的改变.[方法]睾丸匀浆用2,4-二硝基苯肼显色法测定丙酮酸的生成量[1],计算每克睾丸湿组织LDH总活性和LDH-X同工酶活性.[结果]"劳倦过度、房室不节"肾阳虚造型组LDH总活性显著低于正常对照组(P<0.05.P<0.01),金匮肾气丸治疗组较"劳倦过度、房室不节"肾阳虚造型组则显著升高(P<0.05,P<0.01);LDH-X活性造型组第2周较对照组有下降的趋势,但无统计学意义,第4、第6周下降显著(P<0.01);治疗组较造型组第2周有升高趋势,但无统计学意义,第4、第6周则显著升高(P<0.01).[结论]从精子特异酶LDH-X活性改变,证明了"肾主生殖"有现代科学依据,该模型系肾阳虚模型.

Abstract:[Objective] The changes of LDH and LDH-X activity in testes of mice with deficiency of kidney-yang induced by overstrain and intemperate sexual intercourse were examined.[Methods] The contents of pyruvate in testis homogenates was measured and the total LDH activity and LDH-X isoenzyme activity per gram of wet tissue were determined by 2,4-dinitrophenylhydrazine color test in different groups.[Results] The total LDH activity in model group of deficiency of kidney-yang was significantly lower than that in normal control group(P<0.05,P<0.01 respectively)while it was higher in therapy group with Jingui Shenqi pill P<0.05,P<0.01)compared with that in model group.LDH-X activity in the second week in model group was decreased compared with that in control group,but there is no statistical difference.Until the fourth and sixth week the decrease became significant(P<0.05);Compared with LDH-X activity in model group,it was raised in the second week in therapy group,but statistics showed no significance.But it was increased significantly in the fourth and sixth week(P<0.01).[Conclusions] The ground of modern science of "the kidney dominating reproduction" is proved by the changes of LDH-X(specific enzyme of sperm) activity and this model is the one with deficiency of kidney-yang.
