蝎毒活性多肽对大鼠脑缺血-再灌注后IL-1B蛋白表达和神经功能的影响

摘    要:目的:研究蝎毒活性多肽对大鼠脑缺血-再灌注损伤的保护作用,并观察该保护作用对白细胞介素-1B(interleukin- 1B,IL-1B)蛋白表达的影响。方法:采用左侧大脑中动脉插入栓线(Middle cerebral artery occlusion,MCAO)方法制作大鼠脑缺血-再灌注模型,观察蝎毒活性多肽对脑缺血大鼠神经功能的影响;用免疫组化方法标记各组大鼠脑组织海马中IL-1B阳性细胞数量。结果:蝎毒活性多肽可降低大鼠局灶性脑缺血再灌注损伤行为学评分,改善因缺血所致神经行为功能缺失,且随着剂量的增加效应增强,并可以显著减少海马IL-1B阳性细胞的数量(P<0.05)。结论:蝎毒活性多肽对脑缺血损伤有一定的保护作用,且这种作用与其减少脑缺血早期IL-1B表达有关。

Abstract:Objective: To study the protective effect of Scorpion venom active peptides(SVAPs) antagonist focal cerebral ischemia-reperfusion injury in rats and the expression of IL-1B protein and its implication in MCAO rats. Method: The reversible middle cerebral artery occlusion (MCAO) model was established in rats to investigate the role of SVAPs in ischemia injury. The scores of neurological deficits were determined by Lonna EZ method in MCAO rats. The IL-1B protein positive neurons revealed by immunohistochemistry were counted in the PC and Cpu areas. Result: The scores of neurological deficits and the count of IL-1 B protein positive neurons were markedly decreased by SVAPs in a dose- dependent manner when the dosage of SVAPs was increased (P < 0.01). Conclusion: SVAPs can protect MCAO rats antagonist cerebral ischemia- reperfusion injury, the protective effect is related to the expression of IL-1B.
